Hypomethylating agents (HMA) for the treatment of acute myeloid leukemia and myelodysplastic syndromes: mechanisms of resistance and novel HMA-based therapies

DNA hypomethylating agents can reverse DNA methylation and re-program tumor cells, but long-term use leads to resistance. First-generation HMAs are now standard for treating acute myeloid leukemia and myelodysplastic syndromes, but resistance is a growing concern.

Summary: This study provides insights into the DNA methylation patterns in lung tissues of ACD/MPV patients, revealing differential methylation regions that may be involved in the pathogenesis of the disorder. Analysis of patients with FOXF1 mutations or deletions suggests that hypermethylation in the FOXF1 enhancer region may contribute to FOXF1 haploinsufficiency. Additionally, abnormal hypermethylation in exon 1 of FOXF1 may play a role in some cases of ACD/MPV in patients without known pathogenic FOXF1 variations.
