Investigating the inhibitory and penetrating properties of three novel anticancer and antimicrobial scorpion peptides via molecular docking and molecular dynamic simulation

Summary: Bladder cancer, including muscle invasive and non-muscle invasive types, is a common worldwide cancer. Non-muscle invasive bladder cancer requires more therapy due to its higher recurrence rate and longer, more expensive care. Experimental studies have shown that crude scorpion venom and purified proteins and peptides can suppress cancer metastasis, inhibit cancer growth, stop cell cycle, and induce cell apoptosis. In this research, three novel peptides (P2, P3, P4) were discovered from scorpions and studied for their antimicrobial and anticancer properties. Peptides 2 and 3 showed the highest binding rate and were confirmed to penetrate the membrane of cancer cells and bacterial cells. QSAR analysis suggests that peptides 2 and 3 can act as anti-cancer and antimicrobial peptides.

Enterococcus faecalis alters endo-lysosomal trafficking to replicate and persist within mammalian cells

Summary: This study describes the intracellular survival mechanisms of Enterococcus faecalis, a pathogen frequently found in chronic wounds. The researchers found that E. faecalis can survive and replicate inside immune cells and non-immune cells during wound infection, potentially contributing to chronic infections. The bacteria are taken up by host cells primarily through macropinocytosis and manipulate the endosomal pathway to avoid lysosomal degradation and death. The findings provide insights into E. faecalis pathogenesis and highlight the diversity of mechanisms bacteria use to evade host-mediated killing.

Antimicrobial Peptides: A Potent Alternative to Antibiotics

They show various activity spectra and mechanisms of actions, and can be used synergistically against ESKAPE bacteria, particularly in biofilm treatments. Challenges and limitations in their therapeutic applications are highlighted, with potential solutions being evaluated to determine if antimicrobial peptides could potentially replace antibiotics in the future.
