A Novel Compound Heterozygous Gene Mutation of Dolichol Kinase Deficiency (DOLK-CDG)

Shufeng Yu et al.

Summary: The study reports a case of a child with DOLK-CDG diagnosed and treated in the Affiliated Hospital of Qingdao University. The child presented with neonatal asphyxia, Ichthyoid rash, and congenital heart disease. A novel mutation in the DOLK gene was identified before the child's death. A literature review was conducted to summarize the mutation sites and clinical manifestations of DOLK-CDG reported up to August 2021.

Protein O-mannosylation across kingdoms and related diseases: From glycobiology to glycopathology

Jing Hang et al.

Summary: The post-translational glycosylation of proteins by O-linked alpha-mannose is a conserved process found in various organisms. O-mannosylated glycoproteins play important roles in biological processes, and their aberrant glycosylation is associated with disease. O-mannosylation also has diverse functions in lower eukaryotes and prokaryotes. Furthermore, the relationship between O-mannosylation and gut microbiota is highlighted.

Whole-Exome Sequencing Revealed New Candidate Genes for Human Dilated Cardiomyopathy

Ylenia D'Agostino et al.

Summary: Dilated cardiomyopathy (DCM) is a complex disease with genetic and environmental factors contributing to its development. Early identification of causative gene mutations is important for genetic diagnosis, pre-symptomatic interventions, and prognosis prediction. Whole-exome sequencing (WES) has emerged as a promising method to identify causal variants in known genes and novel disease-related candidates in DCM.

Clinical, biochemical and molecular phenotype of congenital disorders of glycosylation: long-term follow-up

Anna Bogdanska et al.

Summary: This study describes the clinical, biochemical, and molecular findings of CDG patients and presents long-term follow-up results. Multiple serum Tf isoform measurements and statistical analysis reveal differences among CDG patients. Mannose supplementation in MPI-CDG patients and galactose supplementation in PGM1-CDG patients show improvement in clinical status and Tf isoform profiles.

Dystroglycanopathy: From Elucidation of Molecular and Pathological Mechanisms to Development of Treatment Methods

Motoi Kanagawa

Summary: Dystroglycanopathy is a group of muscular dystrophies caused by abnormal glycosylation of dystroglycan, with diverse clinical symptoms ranging from congenital to adult-onset types. Research in the 2010s has identified the sugar chain structure and functions of causative gene products, created various model mice for studying pathological mechanisms, and proposed treatment strategies based on glycosylation.

Fatal Neonatal DOLK-CDG as a Rare Form of Syndromic Ichthyosis

Katalin Komlosi et al.

Summary: The cases highlight the phenotypic variability of glycosylation disorders, emphasizing the importance of broadening the differential diagnosis of ichthyosis and actively searching for organ involvement in neonates with ichthyosis. The report describes two neonatal cases of lethal ichthyosis from the same family, caused by compound heterozygous variants in the DOLK gene.

Congenital Disorders of Glycosylation: What Clinicians Need to Know?

Patryk Lipinski et al.

Summary: Congenital disorders of glycosylation (CDG) are clinically heterogeneous disorders with defects in glycans synthesis and attachment. Diagnostic methods include isoelectric focusing of serum transferrin. Next-generation sequencing has improved diagnostics and led to more reported CDG subtypes. Early and accurate diagnosis of CDG is crucial for optimal therapeutic interventions.
