Integrate mechanistic evidence from new approach methodologies (NAMs) into a read-across assessment to characterise trends in shared mode of action

Sylvia E. Escher et al.

Summary: Read-across approaches may not provide sufficient evidence on a common mode of action across category members. A case study on branched aliphatic carboxylic acids shows the potential to induce hepatic steatosis. By analyzing gene expression patterns and adverse outcome pathways, researchers were able to confirm biological similarity and design an in vitro testing battery to systematically investigate a common mode of action among the compounds.

The onset of active gill respiration in post-embryonic zebrafish (Danio rerio) larvae triggers an increased sensitivity to neurotoxic compounds

Nadine Kaemmer et al.

Summary: The study investigates the sensitivity of zebrafish to neurotoxic substances and the impact of swim bladder deflation on respiratory failure syndrome. The results show that post-embryonic 12-day-old larvae are more sensitive than embryos and adult fish, with higher accumulation of substances in 4-day-old fish, and swim bladder deflation may contribute to oxygen deprivation in respiratory failure syndrome.

Beyond the behavioural phenotype: Uncovering mechanistic foundations in aquatic eco-neurotoxicology

Donald Wlodkowic et al.

Summary: In recent years, there has been increased awareness of the impact of anthropogenic pollution on the behavior of aquatic organisms. Behavioral ecotoxicology and eco-neurotoxicology have become popular fields of research, but there is still a lack of understanding in the underlying mechanisms of behavioral alterations. This article discusses the importance of behavioral phenotyping in uncovering the fundamental mechanisms of neurotoxic and neuromodulating contaminants, and highlights the potential applications of neurobiology and neurophysiology analytical strategies.

Worst-case ranking of organic chemicals detected in groundwaters an surface waters in England

David Spurgeon et al.

Summary: The Environment Agency has been using GC-MS and LC-MS to measure organic substances in water since 2009 and 2014 respectively. They use this data to rank the detected substances based on worst-case risk assessment. The approach identified both known and novel substances for further assessment.

Summary: A rapid zebrafish behavioral profiling assay was developed to quantify the neurodevelopmental effects of environmental substances and provide additional information. The assay successfully captured the impacts of different neuroactive substances on spontaneous movement. Additionally, the web-based tool EMAnalysis was provided to accelerate the discovery of neurotoxic environmental contaminants.

Tail Coiling Assay in Zebrafish (Danio rerio) Embryos: Stage of Development, Promising Positive Control Candidates, and Selection of an Appropriate Organic Solvent for Screening of Developmental Neurotoxicity (DNT)

Andreia A. S. de Oliveira et al.

Summary: The tail coiling assay in zebrafish embryos is proposed as an alternative method for screening developmental neurotoxicity induced by chemicals. Embryos from 22 to 29 hpf showed consistent coiling activity, with caffeine and MS-222 affecting activity levels differently. Using 1% DMSO as a solvent for the assay was considered relatively safe.

Neurotoxicity and underlying cellular changes of 21 mitochondrial respiratory chain inhibitors

Johannes Delp et al.

Summary: The study utilized 21 pesticides described as mitochondrial inhibitors to investigate the adverse outcome pathway (AOP:3) of Parkinson's disease, finding that some inhibitors triggered neurodegeneration while others did not. There was a clear hierarchy of assay sensitivity, with mitochondrial failure being much more sensitive than neurite damage in predicting toxic reactions.

Cytochrome P450-dependent biotransformation capacities in embryonic, juvenile and adult stages of zebrafish (Danio rerio)-a state-of-the-art review

Ann-Kathrin Loerracher et al.

Summary: There is a strong trend towards using zebrafish embryos as translational models in ecotoxicological and toxicological testing strategies, leading to an increasing need to understand their biotransformation capacity. However, there is still insufficient research on the expression patterns and activities of CYP isoforms in juvenile and adult zebrafish. Studies have shown qualitative similarities but quantitative differences in metabolic activities between zebrafish embryos and later developmental stages, highlighting the importance of further research to assess metabolic profiles and capacities of CYP isoforms in zebrafish.

TCMacro: A Simple and Robust ImageJ-Based Method for Automated Measurement of Tail Coiling Activity in Zebrafish

Kevin Adi Kurnia et al.

Summary: The TCMacro method developed in this study provides a simple and robust way to measure relative tail coiling activities in zebrafish embryos in a high throughput manner. The method is based on dynamic pixel intensity changes in a region of interest, allowing for accurate and batch measurements of tail coiling counts, durations and intervals. The method was successfully validated through neurotoxicity analysis of Tricaine and caffeine, showing dose-dependent decrease and increase in tail coiling activity, respectively.

Automated measurement of the spontaneous tail coiling of zebrafish embryos as a sensitive behavior endpoint using a workflow in KNIME

Afolarin O. Ogungbemi et al.

Summary: Neuroactive substances are the largest group of chemicals detected in European surface waters and can induce adverse neurological effects in humans and organisms. Developing new screening tools based on the 3R principle is advocated by the scientific community to assess neurotoxicity without inflicting pain and stress on animals. Using behavior measurement of zebrafish embryos as an alternative to adult animal testing is considered as a valid and valuable method.

Translating Neurobehavioral Toxicity Across Species From Zebrafish to Rats to Humans: Implications for Risk Assessment

Charles V. Vorhees et al.

Summary: Neurotoxicological science can be studied through a spectrum of approaches, each with its own advantages and limitations. Experimental animal models provide cause-and-effect information, human epidemiological and clinical studies give information closest to human risk analysis, and in vitro methods offer high throughput. Cross-species translation is crucial for risk assessment and mechanistic determination.
