Palladium-Catalyzed Alkenyl C-H Activation/Diamination toward Tetrahydrocarbazole and Analogs Using Hydroxylamines as Single- Nitrogen Sources

Summary: A palladium-catalyzed alkenyl C-H activation/diamination reaction of cycloalkenyl bromoarenes with hydroxyl-amines is presented. A wide range of tetrahydrocarbazoles and analogs were synthesized using well-tuned bifunctional secondary hydroxylamines as the sole nitrogen sources. Mechanistic investigations indicate that the selective alkenyl C-H activation/diamination cascade process is responsible for the formation of N-heterocycles.

Catellani Reaction: An Enabling Technology for Vicinal Functionalization of Aryl Halides by Palladium(0)/Norbornene Cooperative Catalysis

Summary: The Catellani reaction, discovered by Catellani in 1997 and further developed by Catellani, Lautens, and others, is a powerful strategy for the synthesis of polysubstituted arenes. This method allows for the simultaneous functionalization of both ortho- and ipso-positions of aryl halides with different electrophiles and terminating agents under the cooperative catalysis of palladium and norbornene. The review focuses on the significant progress in such transformations and categorizes typical Catellani reactions into five parts based on the functionalization modes of ortho-C-H bonds: alkylation, arylation, amination, acylation, or thiolation.

Origin of Ligand Effects on Stereoinversion in Pd-Catalyzed Synthesis of Tetrasubstituted Olefins

Summary: DFT calculations and EDA were used to investigate the mechanism and origin of ligand effects on stereoinversion in Pd-catalyzed synthesis of tetrasubstituted olefins. It was found that the steric properties of the ligands lead to different stereoselectivity-determining steps, with Pauli repulsion and polarization playing dominant roles in controlling stereochemistry in 1,2- and 1,3-migrations, respectively. Differences in Pauli repulsion and polarization between the two stereoselective transition states were further identified.

N-Tosyloxyphthalimide serves as an alternative amidation reagent, allowing for high efficiency and good functional group tolerance. The reaction can be scaled up to gram scale and the amidated product can be easily converted into valuable scaffolds with one-step derivatization.

Palladium-Catalyzed Sequential C-H Activation/Amination with Diaziridinone: An Approach to Indoles

Possible coordination of an installed terminal alkenyl group with palladium and steric hindrance were proposed to be responsible for the monoamination selectivity.
