A single-cell transcriptomic atlas characterizes cell types and their molecular features in yak ovarian cortex

Jie Pei et al.

Summary: Single-cell RNA sequencing was used to investigate the molecular characteristics of various cell types in the yak ovarian cortex, and a cellular atlas was successfully constructed. Differential gene expression analysis identified differentially expressed genes in endothelial cells, immune cells, stromal cells, smooth muscle cells, oocytes, macrophages, epithelial cells, and granulosa cells. The cellular atlas revealed the biological characteristics of the ovarian cortex at the cellular molecular level and provided insights into female reproductive biology via cellular communications in the yak.

Single-cell analysis of human testis aging and correlation with elevated body mass index

Xichen Nie et al.

Summary: This study utilized single-cell RNA-seq to investigate the effects of aging on testicular cells in men. The results revealed age-related changes in multiple cell types, including alterations in signaling pathways, inflammation, and metabolism. The extent of dysregulation was also found to be correlated with body mass index in older men. This research provides insights into the candidate molecular mechanisms underlying testicular changes caused by aging.

Identification of the TSSK4 Alternative Spliceosomes and Analysis of the Function of the TSSK4 Protein in Yak (Bos grunniens)

Xingdong Wang et al.

Summary: In mammals, the testis-specific serine/threonine kinase (TSSK) is crucial for spermatogenesis and male fertility. This study successfully cloned multiple alternative spliceosomes of the TSSK4 gene in yak testis tissue, providing a foundation for future investigations into the function of each spliceosome.

Summary: The study investigated the expression and localization of Hsp60 gene in testicular development of cattle-yak and yak. Results showed variations in Hsp60 expression among different tissues and developmental stages, with significantly higher expression in cattle-yak than yak. Hsp60 proteins were found to be located in epithelial cells, spermatocytes, sperm cells, and mesenchymal cells.

Summary: This study provides insights into the complex changes in cell identity during human testis development in prenatal life, from the establishment of testicular cords to the transitioning of male primordial germ cells into cells resembling f0 spermatogonia. The analysis of single-cell transcriptomes reveals the coordinated and temporal development of embryonic, fetal, and postnatal male germline and somatic niche.

Summary: The study focused on the mitochondrial genome of yak populations from Qinghai and Xinjiang, revealing a high mitochondrial diversity and three distinct branches. The analysis identified a new phylogenetic branch containing both wild and domestic yaks. Mitotype clustering divided the 155 mitotypes found in 206 individuals into 3 haplogroups, not associated with the geographical distribution of yaks.

Summary: This study characterized the transcriptomes of various cell types in porcine testes using scRNA-seq, identifying different subsets of spermatogonia and novel cell surface markers. Functional enrichment analysis revealed the diverse roles these cells play in spermatogenesis. The data provides valuable insights into porcine spermatogenesis and lays the foundation for identifying key molecular markers in the development of male germ cells.

Characterization of N6-Methyladenosine in Domesticated Yak Testes Before and After Sexual Maturity

Xingdong Wang et al.

Summary: The study demonstrates that m(6)A methylation levels in yak testes increase after sexual maturation, with differential methylation peaks observed in different stages of maturity, primarily concentrated in the exon region. KEGG pathway analysis suggests involvement of homologous recombination, Notch signaling pathway, growth hormone synthesis, and other signaling pathways in testicular development and maturation in yaks.
