Interaction of TOR and PKA Signaling in S. cerevisiae

Michael Plank

Summary: TOR and PKA signaling pathways play important roles in growth regulation and nutrient sensing in Saccharomyces cerevisiae. This review explores the association between these pathways, focusing on shared target proteins. It discusses the potential direct regulation and cross-talk between TOR and PKA, as well as the transcriptional responses elicited by these pathways.

Fast detection of PKA activity in Saccharomyces cerevisiae cell population using AKAR fluorescence resonance energy transfer probes

Sonia Colombo et al.

Summary: In this study, AKAR3 and pm-AKAR3 probes were used with a plate reader to monitor PKA activity in Saccharomyces cerevisiae cells. Results showed that glucose addition increased PKA activity, and this change depended on the presence and strength of PKA. Furthermore, the pm-AKAR3 probe allowed monitoring of PKA activity from the starting site to internal regions.

Cellpose 2.0: how to train your own model

Marius Pachitariu et al.

Summary: This study introduces Cellpose 2.0, a software package that includes diverse pretrained models and a human-in-the-loop pipeline for rapid customization of models. The results show that models pretrained on the Cellpose dataset can be fine-tuned with fewer user annotations and still achieve high-quality segmentations.

A yeast FRET biosensor enlightens cAMP signaling

Dennis Botman et al.

Summary: The cAMP-PKA signaling cascade in budding yeast regulates adaptation to changing environments through a combination of extracellular sensing and intracellular metabolism, with the cAMP peak height conveying information about prospective growth rates according to the Weber-Fechner law. The yEPAC biosensor provides a new avenue for understanding yeast physiology, signaling, and metabolic adaptation.

Nutrient sensing and cAMP signaling in yeast: G-protein coupled receptor versus transceptor activation of PKA

Griet Van Zeebroeck et al.

Summary: The study investigates the PKA pathway in regulating cell growth in yeast, showing different nutrient activation mechanisms, some mediated by cAMP and others through direct release of catalytic subunits to activate PKA. Nutrient transceptors may be evolutionary ancestors of GPCR, using a more primitive direct signaling mechanism compared to the indirect cAMP signaling of GPCRs.
