Regulation of Translation, Translocation, and Degradation of Proteins at the Membrane of the Endoplasmic Reticulum

Lea Daverkausen-Fischer et al.

Summary: The endoplasmic reticulum (ER) plays a central role in protein folding and secretion. BiP and ERdj proteins are important chaperones in the ER, assisting in the folding process and regulating protein synthesis, translocation, and degradation. Dysfunction of these proteins can lead to the accumulation of unfolded proteins and the development of metabolic diseases.

Osmoregulation and the Hypothalamic Supraoptic Nucleus: From Genes to Functions

Andre Souza Mecawi et al.

Summary: Water molecules equilibrate the potential gradient between intra and extracellular compartments due to the high permeability of the plasma membrane; Osmoregulatory mechanisms have evolved to maintain strict limits on the tonicity of extracellular fluid; Studies focus on key concepts and mechanisms in adjusting the internal environment to cope with challenges of water imbalance.

Transcription factor Creb3l1 maintains proteostasis in neuroendocrine cells

Mingkwan Greenwood et al.

Summary: This study aims to explore the function and regulatory mechanism of Creb3l1 in hypothalamic neurons. Through a multiomic strategy, it is found that Creb3l1 plays a crucial role in the regulation of ribosomal proteins and endoplasmic reticulum proteins. Knockdown of Creb3l1 significantly reduces neuropeptide synthesis and secretion, leading to changes in food and water consumption. This research provides a new therapeutic strategy for metabolic diseases.

Single-cell analysis of the aging female mouse hypothalamus

Kaitlyn H. Hajdarovic et al.

Summary: This study investigates the cell-specific changes in the aging hypothalamus through single-nuclei RNA sequencing. The authors identify age-related features in neuronal subtypes, as well as astrocytes and microglia. Interestingly, elevated expression of the X inactivation gene Xist is observed in hypothalamic neurons of aging female mice. Machine learning analysis shows that the levels of X chromosome genes and Xist can accurately predict cellular age.

Transcription factor Creb3l1 maintains proteostasis in neuroendocrine cells

Mingkwan Greenwood et al.

Summary: By investigating the role of Creb3l1 in the hypothalamus, this study revealed its comprehensive control over the PERK signaling pathway in magnocellular neurones, impacting neuropeptide synthesis and secretion, which in turn influence food and water consumption.

Summary: The idea that water intake or hydration might have an intrinsic, independent role in modulating the risk of metabolic diseases is relatively new. Higher levels of AVP are associated with an increased risk of developing type 2 diabetes and components of the metabolic syndrome.
