Iron in immune cell function and host defense

David Haschka et al.

Summary: Iron plays a crucial role in the interaction between host, pathogen, and immune responses. Restricting iron availability to invading pathogens is a powerful defense mechanism known as nutritional immunity. Iron also influences immune cell function and differentiation, as well as inflammation control.

Anti-Ro60 and anti-Ro52/TRIM21: Two distinct autoantibodies in systemic autoimmune diseases

Adrian Y. S. Lee et al.

Summary: The diagnostic autoantibodies anti-Ro60 and anti-Ro52/TRIM21 are commonly found in systemic autoimmune disorders like SLE, often co-existing despite lacking physical linkage, signaling a convergence of failed immunological tolerance processes. Sometimes classified together as the SSA or Ro autoantibody system without clear distinction, further research is needed to explore separate detection and reporting for these antibodies and their roles in autoimmunity. Understanding their origins and pathogenic functions will provide insights for preventing and treating autoimmune disorders.

Iron robbery by intracellular pathogen via bacterial effector-induced ferritinophagy

Qi Yan et al.

Summary: Etf-3 is a bacterial protein that induces ferritinophagy to facilitate pathogen iron capture, allowing the bacterium to acquire iron for survival and proliferation in the host cell.

Host immunity increases Mycobacterium tuberculosis reliance on cytochrome bd oxidase

Yi Cai et al.

Summary: Tuberculosis, caused by Mycobacterium tuberculosis (Mtb), is a major global health problem resulting in over one million deaths annually. Mtb's ability to adapt to various immunological and environmental pressures, including through flexible respiratory capacity, is crucial for its pathogenic success. This study highlights the importance of cytochrome bd oxidase in Mtb's resistance to the adaptive immune response, particularly in acidic conditions induced by IFN gamma, and shows its impact on the efficacy of a potential new tuberculosis therapy.

Hinokitiol chelates intracellular iron to retard fungal growth by disturbing mitochondrial respiration

Xueyang Jin et al.

Summary: The study aimed to investigate the potential use of hinokitiol as an iron chelator for treating fungal infections. Results showed that hinokitiol selectively chelated iron in fungal cells, inhibited fungal cell respiration, and had minor effects on mammalian cells. Hinokitiol also demonstrated low potential for inducing resistance in Candida species and improved survival rates in a Galleria mellonella infection model.

Ferritin H deficiency deteriorates cellular iron handling and worsens Salmonella typhimurium infection by triggering hyperinflammation

David Haschka et al.

Summary: Iron is essential for both mammals and pathogens, and inflammation-induced changes in iron metabolism play a crucial role in host defense against infections. The iron storage protein FTH in myeloid cells is important for controlling bacterial spread and modulating NF-κB and inflammasome-mediated cytokine activation.
