Recent advances in the biosynthesis of ribosomally synthesized and posttranslationally modified peptides of fungal origin

Summary: Ribosomally synthesized and posttranslationally modified peptides (RiPPs) are a class of natural products with potent biological activities, and remarkable structural diversity is generated through posttranslational modifications (PTMs). Fungal RiPPs (F-RiPPs) have fewer examples, but recent progress in sequencing of fungal genomes indicates hidden RiPP pathways in fungal resources, making F-RiPPs an attractive target for genome mining studies.

Oxidative bicyclic ring system formation involving indole diterpene biosynthesis: Remarkable substrate tolerance of a prenyltransferase and flavoprotein oxidase

Summary: Oxidative modifications are essential for the biosynthesis of indole diterpenes (IDTs), and this study focused on the role of prenyltransferases and flavoprotein oxidases in the synthesis of nodulisporic acid-type and shearinine-type bicyclic rings on the indole moiety. The enzymatic reaction of prenyltransferase JanD showed that it recognizes a C13 hydroxy group on the diterpene moiety. Flavoprotein oxidases JanO and NodO exhibited different substrate specificities for penta/hexacyclic IDTs and pentacyclic IDTs, respectively.

Heterologous expression of a polyketide synthase ACRTS2 in Aspergillus oryzae produces host-selective ACR toxins: coproduction of minor metabolites

Summary: by heterologously expressing the ACRTS2 gene, we successfully produced the host-selective ACR toxin for brown leaf spot on rough lemon. Minor ACR-toxin-related metabolites were discovered in the process. Further investigations led to the determination of a decarboxylation mechanism and a novel branching route for the formation of byproducts. The involvement of putative cyclase ACRTS3 was excluded. The production of the ACR toxin in AO is solely responsible for ACRTS2.

Molecular and Computational Bases for Spirofuranone Formation in Setosusin Biosynthesis

Summary: This study identified the biosynthetic gene cluster of Setosusin in the fungus Aspergillus duricaulis and elucidated its biosynthetic pathway through enzyme reconstitution experiments. The crucial role of the cytochrome P450 enzyme SetF in spirofuranone synthesis was revealed, showing its involvement in epoxidation and structural rearrangement processes. Mutagenesis experiments identified Lys303 as a potential catalytic residue important for spirofuranone synthesis.

Biochemistry-Guided Prediction of the Absolute Configuration of Fungal Reduced Polyketides

Summary: Highly reducing polyketide synthases (HR-PKSs) are responsible for the production of structurally diverse polyketides (PKs) through a complex stereochemical process. By examining the synthesis of polyhydroxy PKs, researchers were able to determine the absolute configurations of stereogenic centers using chemical degradation and comparison with synthetic standards. This study proposes a stereochemical rule to explain the absolute configurations of reduced PKs and highlights discrepancies in previously reported structures.

Cytochrome P450 enzymes in fungal natural product biosynthesis

Cytochrome P450 enzymes play essential roles in fungal natural product biosynthesis with broad substrate scope and high catalytic versatility. Recent technical advances, such as efficient genome editing techniques and development of filamentous fungal strains as P450 expression hosts, have led to remarkable achievements in fungal P450 studies.
