Prognostic validation of a new classification system for extent of resection in glioblastoma: A report of the RANO resect group

Philipp Karschnia et al.

Summary: This study investigated the prognostic value of the extent of resection in glioblastoma, finding that smaller volumes of residual contrast-enhancing tumor were associated with better outcomes. Additionally, removing non-contrast-enhancing tumor beyond the tumor borders resulted in improved survival. These findings have significant implications for stratification in clinical trials.

Differential expression of checkpoint markers in the normoxic and hypoxic microenvironment of glioblastomas

Stine Asferg Petterson et al.

Summary: Glioblastoma, the most common primary malignant brain tumor, has a low overall survival rate of 14.6 months. This study investigated the expression of immune-related proteins in hypoxic tumor areas and found that CD44 and B7-H3 were upregulated while VISTA was downregulated. These heterogeneous expressions have implications for the development of novel therapeutic strategies.

T-cell dysfunction in the glioblastoma microenvironment is mediated by myeloid cells releasing interleukin-10

Vidhya M. Ravi et al.

Summary: The study demonstrates that a subset of myeloid cells in Glioblastomas induce dysfunctional T cells through Interleukin-10 signaling, and inhibiting downstream JAK/STAT pathway could restore the responsiveness of immune therapy. The findings are validated using in vitro and in vivo models, providing further evidence of Interleukin-10 release being a driving force of tumor immune escape. Integrative modeling of single cell and spatial transcriptomics data proves to be a valuable tool for studying the tumor immune microenvironment and may contribute to the development of successful immunotherapies.

Spatially resolved multi-omics deciphers bidirectional tumor-host interdependence in glioblastoma

Vidhya M. Ravi et al.

Summary: This study utilizes spatially resolved transcriptomics, metabolomics, and proteomics to characterize glioblastomas and uncover their spatial organization and adaptive mechanisms to inflammatory and metabolic stimuli. The integration of metabolic imaging and imaging mass cytometry reveals the interdependence between tumor and host in a locoregional manner, leading to spatially exclusive adaptive transcriptional programs. The findings confirm the importance of environmental stress in shaping glioblastoma subclones and their transcriptional programs.

Exploring the Impact of TREM2 in Tumor-Associated Macrophages

Darya Khantakova et al.

Summary: TREM2 is a receptor expressed in tumor-associated macrophages and is associated with tumor control and the tumor immune environment. Blocking or targeting TREM2 can improve tumor control, enhance immune response, and alter the tumor immune environment. Preclinical studies have paved the way for further investigation into TREM2 targeting therapies.

Leveraging extrachromosomal DNA to fine-tune trials of targeted therapy for glioblastoma: opportunities and challenges

Imran Noorani et al.

Summary: Intratumour heterogeneity in glioblastoma poses challenges for effective treatment. The amplification of oncogenes on extrachromosomal DNA (ecDNA) plays a key role in accelerating tumour evolution and resistance. Current targeted therapies have failed to improve outcomes in glioblastoma. Detection of ecDNA can optimize patient selection for clinical trials and enhance treatment efficacy.

Tumor cell and immune cell profiles in primary human glioblastoma: Impact on patient outcome

Maria Gonzalez-Tablas Pimenta et al.

Summary: This study investigated the cellular composition of 55 GBM samples and correlated the tumor immune profile with patient features and outcomes. The GBM samples primarily consisted of tumor and normal astrocytic cells, along with a significant but variable fraction of immune cells. Three distinct immune profiles were identified in the GBM samples, with untreated patients showing worse outcomes when immune infiltrates included a mixed population of myeloid and T-lymphoid cells.

Glioblastomas acquire myeloid-affiliated transcriptional programs via epigenetic immunoediting to elicit immune evasion

Ester Gangoso et al.

Summary: In this study, it was found that GBM stem cells acquire the ability to evade immune clearance through epigenetic immunoediting, leading to the formation of an immunosuppressive tumor microenvironment. This process is not driven by genetic selection but by altering the transcriptional and epigenetic profile of GSCs after immune attack.

Interactions between cancer cells and immune cells drive transitions to mesenchymal-like states in glioblastoma

Toshiro Hara et al.

Summary: The mesenchymal subtype of glioblastoma is believed to be influenced by both cancer cell-intrinsic alterations and external cellular interactions, with macrophages inducing a transition of glioblastoma cells into mesenchymal-like states. This transition is mediated by macrophage-derived oncostatin M (OSM) interacting with its receptors on glioblastoma cells, leading to the activation of STAT3. This study also reveals that mesenchymal-like glioblastoma states are associated with increased expression of a mesenchymal program in macrophages and enhanced cytotoxicity of T cells, suggesting potential therapeutic implications.
