Detection of singlet oxygen by EPR: The instability of the nitroxyl radicals

Henrique F. V. Victoria et al.

Summary: The use of spin traps and redox probes coupled with electron paramagnetic resonance (EPR) is an effective method in evaluating the efficiency of photosensitizers and photocatalysts in various processes involving reactive oxygen species. This study analyzed the kinetics of radical generated by the oxidation products of a specific redox probe (Hydroxy-TEMP) in the presence of water-soluble porphyrins under white light illumination. Factors such as probe concentration, pH, and porphyrin photostability were evaluated, and the mechanism of radical degradation and porphyrin photobleaching were investigated. The results provide valuable insights into the evaluation and interpretation of photosensitizer efficiency.

Surface Tension and Self-association Properties of Aqueous Polysorbate 20 HP and 80 HP Solutions: Insights into Protein Stabilisation Mechanisms

Patrick Garidel et al.

Summary: The study focused on determining the self-association properties of compendial grade polysorbate 20 HP (PS 20 HP) and polysorbate 80 HP (PS 80 HP) regarding micelle size, critical micelle concentration range (cmr), and surface tension. Results showed that PS 80 HP formed larger micelles compared to PS 20 HP, with critical micelle concentration range values (cmr) at 25 degrees Celsius being considerably lower for PS 80 HP. The temperature dependency of the micelle size was more pronounced for PS 80 HP.

Investigating photodegradation of antibodies governed by the light dosage

Wolfgang Kaiser et al.

Summary: This study investigated the photodegradation of three monoclonal antibodies under visible light. The results show that visible light can induce the formation of various oxidation-related species in the antibodies. The rate of degradation is dependent on light intensity, but the yield is correlated to the applied light dosage, following the Bunsen-Roscoe reciprocity law.

Minimizing Oxidation of Freeze-Dried Monoclonal Antibodies in Polymeric Vials Using a Smart Packaging Approach

Nicole Haerdter et al.

Summary: Primary containers made of cyclic olefin polymer (COP) offer advantages such as high break resistance and biocompatibility, but they are more permeable for gases, which can lead to oxygen ingress. The use of smart packaging, including oxygen and moisture absorbers, can help reduce oxidation and control residual moisture, potentially providing a promising approach for ensuring the long-term stability of biopharmaceutical products.

Pharmaceutical Excipients Enhance Iron-Dependent Photo-Degradation in Pharmaceutical Buffers by near UV and Visible Light: Tyrosine Modification by Reactions of the Antioxidant Methionine in Citrate Buffer

Natalia Subelzu et al.

Summary: The study found that sugars and amino acids significantly promote the oxidation of peptide Met, without increasing the yields of hydrogen peroxide. The addition of free Met reduces the oxidation of peptide Met, but may cause the addition of Met oxidation products to Tyr residues of the target peptides in citrate buffer. Overall, commonly used excipients enhance the light-induced oxidation of amino acids in model peptides.
