Concurrent de novo ZFHX4 variant and 16q24.1 deletion in a patient with orofacial clefting; a potential role of ZFHX4 and USP10

Summary: This study describes a girl with multiple craniofacial anomalies including cleft lip, alveolus, and palate, tooth agenesis, and dysmorphic features. Various genetic diagnostic tests were performed, including cleft gene panel analysis, SNP array, WGS, WES, and Q-PCR analysis. The results identified a maternal deletion at 16q24.1 encompassing the candidate gene USP10 and a de novo Loss-of-Function variant in the ZFHX4 gene. The mRNA expression of certain putative target genes involved in orofacial clefting was found to be lowered in the patient compared to controls, suggesting the involvement of ZFHX4 as a novel cleft gene and the potential contribution of USP10 to the etiology of orofacial clefts in humans.

Identification of genomic imbalances in oral clefts
