A [Pt(cis-1,3-diaminocycloalkane)Cl2] analog exhibits hallmarks typical of immunogenic cell death inducers in model cancer cells

Vojtech Novohradsky et al.

Summary: Platinum drugs are commonly used in cancer treatment, but there is active research in developing new metal-based drugs with different mechanisms. The combination of cytotoxic chemotherapy and immunotherapy has shown promising synergistic effects, while the development of metal-based drugs that exhibit cytostatic effects and induce immunogenic cell death is of interest for cancer treatment.

In Situ Assembly of Platinum(II)-Metallopeptide Nanostructures Disrupts Energy Homeostasis and Cellular Metabolism

Zhixuan Zhou et al.

Summary: Nanostructures play essential roles in nature and contribute to the diversity of life. In this study, we designed a platinum-containing tripeptide that forms intracellular fibrillar nanostructures upon rearrangement in the presence of endogenous H2O2. The formed nanostructures block metabolic functions and regulate ATP production, actin formation, and histone deacetylase activity. This research demonstrates the potential of assembly-driven nanomaterials in drug discovery.

Ferroptosis Photoinduced by New Cyclometalated Iridium(III) Complexes and Its Synergism with Apoptosis in Tumor Cell Inhibition

Hao Yuan et al.

Summary: The limited therapeutic efficacy of photodynamic therapy (PDT) to hypoxic solid tumors has been improved by two new benzothiophenylisoquinoline-derived cyclometalated Ir-III complexes, IrL1 and MitoIrL2. Both complexes demonstrated a type I PDT process and induced photoinduced ferroptosis in tumor cells under hypoxia, with MitoIrL2 outperforming IrL1 in inhibiting tumor growth through a synergistic effect of ferroptosis and apoptosis.

Single-Cell Chemistry of Photoactivatable Platinum Anticancer Complexes

Elizabeth M. Bolitho et al.

Summary: The Pt(IV) prodrug Pt2 shows remarkable photocytotoxicity towards cancer cells, inducing significant cellular damage and vacuole formation, along with increased accumulation of Pt species in cells compared to dark conditions. Synchrotron techniques reveal only partial reduction of Pt2 upon irradiation, suggesting that phototoxicity in cancer cells may involve both Pt(II) photoproducts and photoexcited Pt(IV) species.

Intracellular Dynamic Assembly of Deep-Red Emitting Supramolecular Nanostructures Based on the Pt horizontal ellipsis Pt Metallophilic Interaction

Xue-Quan Zhou et al.

Summary: Nanorods self-assembled via the Pt ellipsis Pt interaction can cross the cell membrane and move directly to mitochondria without passing by lysosomes. This study suggests that the interaction is strong enough to generate emissive nanoparticles inside cells, but labile enough to allow the nanostructures to reach the mitochondria.

Oxidative Polymerization in Living Cells

Yiheng Dai et al.

Summary: This study demonstrates a novel oxidative polymerization reaction in cells triggered by intracellular ROS, selectively inducing apoptosis in cancer cells. The polymerization products disrupt intracellular antioxidant systems, leading to increased oxidative stress, further promoting oxidative polymerization and activating apoptosis pathways related to ROS. The anticancer efficacy and biosafety of this strategy are proven in vitro and in vivo, suggesting a new possibility for chemists to manipulate cellular behavior through artificial chemical reactions.
