Antisense Oligonucleotide Therapeutics for Cystic Fibrosis: Recent Developments and Perspectives

Young Jin Kim et al.

Summary: Antisense oligonucleotide (ASO) technology has been successful in developing therapeutic options for diseases, such as Mendelian disorders and cystic fibrosis (CF). ASOs can modulate gene expression and have shown promising results in treating spinal muscular atrophy and CF. This review discusses the mechanisms of ASO therapies, the challenges, and the current state and prospects of ASO therapies for CF.

Emulsions of hydrolyzable oils for the zero-order release of hydrophobic drugs

Laura Tebcharani et al.

Summary: This study introduces a drug delivery system using a gellified emulsion for zero-order release of hydrophobic drugs, demonstrating tunable release rate and period by varying initial oil and drug concentrations. The system's quantitative understanding allows for predicting drug release kinetics, and it remains fully functional even after storing at -20 degrees C. Cell viability studies confirm the non-toxicity of the hydrolyzable oil and its hydrolysis product under the utilized conditions.

Tuning DNA-nanoparticle conjugate properties allows modulation of nuclease activity

Jeff C. Hsiao et al.

Summary: This study systematically investigates the interactions between nuclease enzymes and oligonucleotide-coated gold nanoparticles, revealing new phenomena that can be applied to functional nanosystems. The study also demonstrates that nuclease activity can be tuned by modifying the characteristics of the nanoparticles.
