The Role of NK Cells and Their Exosomes in Graft Versus Host Disease and Graft Versus Leukemia

Alireza Zafarani et al.

Summary: NK cells play an important role in preventing and controlling tumors and viral diseases, but their role in hematopoietic stem cell transplantation is not fully understood. Some research suggests that NK cells can prevent infections and tumor relapse without causing graft versus host disease. NK cell-derived exosomes also have anti-leukemia effects in graft-versus-leukemia. Furthermore, NK cells have antiviral effects in post-HCT cytomegalovirus infection.

Advances in NK cell production

Fang Fang et al.

Summary: Immunotherapy based on natural killer (NK) cells is a promising approach for treating a variety of cancers. Strategies for obtaining large quantities of NK cells with high purity and cytotoxicity have been developed, including using different cellular sources and genetic modification technologies. Closed, automated, and GMP-controlled large-scale expansion systems are also being studied.

NK-cell exhaustion, B-cell exhaustion and T-cell exhaustion-the differences and similarities

Kevin Roe

Summary: T-cell exhaustion has been extensively studied, while less attention has been given to NK-cell exhaustion and B-cell exhaustion. There are differences and similarities between lymphocyte exhaustion, including NK-cell exhaustion, B-cell exhaustion, and T-cell exhaustion. Lymphocyte exhaustion is often confused with anergy, cellular senescence, and suppression, and it is not a binary state but rather a continuum caused by different levels and duration of continuous antigenic stimulation.

Establishment of an efficient ex vivo expansion strategy for human natural killer cells stimulated by defined cytokine cocktail and antibodies against natural killer cell activating receptors

Tsutomu Nakazawa et al.

Summary: This study showed that a robust expansion system for highly purified human NK cells using a defined cytokine cocktail and anti-NK cell activating receptor antibodies was established. The system could be feasible for autologous or allogeneic NK cell-based immunotherapy of GBM and easily controlled under Japanese law on regenerative medicine. The expansion process can enhance NK cell purity and activity for potential cancer treatment.

Temozolomide-induced aplastic anaemia: Case report and review of the literature

Peter J. Gilbar et al.

Summary: The case report describes a 68-year-old female with GBM who developed very severe aplastic anemia (AA) likely due to temozolomide (TMZ) treatment, requiring platelet transfusions, antibiotics, antiviral and antifungal prophylaxis, and G-CSF. The patient's blood counts returned to normal after 38 days, highlighting the importance of monitoring for rare but potentially fatal toxicities associated with TMZ.

NK cell-based cancer immunotherapy: from basic biology to clinical development

Sizhe Liu et al.

Summary: NK cells, as a specialized immune effector cell type, play a critical role in immune activation against abnormal cells. Unlike T cell activation, NK cell activation is governed by interactions between NK receptors and target cells, leading to significant attention in cancer immunotherapy. Many efforts are focused on developing and engineering NK cell-based cancer immunotherapy.

Evaluation of Comprehensive Gene Expression and NK Cell-Mediated Killing in Glioblastoma Cell Line-Derived Spheroids

Takayuki Morimoto et al.

Summary: Glioblastoma (GBM) is the most aggressive primary malignant brain tumor in adults with poor survival rates. Immunotherapy utilizing Natural killer (NK) cells shows promise in treating GBM by exerting a cytotoxic effect on tumor cells. Preclinical evaluation using an ex vivo 3D spheroid model derived from GBM cell lines showed that NK cells had an anti-tumor effect by inducing tumor cell death through apoptosis, providing valuable information for future NK cell-based immunotherapy for GBM patients.

Harnessing features of adaptive NK cells to generate iPSC-derived NK cells for enhanced immunotherapy

Karrune Woan et al.

Summary: Researchers have successfully generated a NK cell subset called iADAPT by editing induced pluripotent stem cells, which exhibited persistence in vivo and superior antitumor activity. This finding suggests that unique subsets of the immune system can be modeled through iPSC technology, providing a new approach for treating patients with advanced cancer.

Yoichi Shida et al.

Summary: GiNKs, as a promising cell-based immunotherapy, show potential in treating GBM by prolonging the survival of NOG mice carrying U87MG-derived tumors. Although PD-L1 is expressed on U87MG and T98G cells, the expression levels vary significantly. PD-1 blocking antibodies did not have an additional effect on prolonging survival.

Mitochondria as Playmakers of CAR T-cell Fate and Longevity

Hosein Rostamian et al.

Summary: The development of CAR-T cell therapy has changed the paradigm of cancer treatment, but patients often do not benefit due to poor cell persistence. Strategies based on long-lasting memory T cells may enhance the therapeutic effects of CAR-T cells. Mitochondrial metabolic pathways play a crucial role in determining the fate, function, and longevity of T cells.

NK Cells in a Tug-of-War With Cancer: The Roles of Transcription Factors and Cytoskeleton

E. Hui Clarissa Lee et al.

Summary: NK cells play a key role in shaping the immune response against cancer, but their functions are hindered by the immunosuppressive tumor microenvironment. While checkpoint inhibition shows promise in early-stage studies, further exploration is needed in the context of the TME. Surface receptors and T-box transcription factors may play crucial roles in identifying and characterizing NK cell dysfunction.
