The Switch and Reciprocating Models for the Function of ABC Multidrug Exporters: Perspectives on Recent Research

Summary: ATP-binding cassette (ABC) transporters are a large group of primary active transporters that couple energy to transport substrates across cellular membranes. Their structure consists of nucleotide binding domains (NBDs) and transmembrane domains (TMDs). Although the NBDs are highly conserved, the TMDs are divergent to accommodate different substrates. The Switch or Alternating Access Model proposes that the NBDs are widely separated and the TMDs and NBDs form an intracellular-facing inverted V shape. In contrast, the Constant Contact model suggests that the NBDs operate in close proximity throughout the transport cycle in an asymmetric allosteric manner. Recent studies have provided insights into these models using nuclear magnetic resonance and mass spectrometry.
