The method showed high capture efficiency and could detect low concentrations of the target gene within a short time. It has potential for the rapid detection of other resistance genes as well.

Hydrophobic magnetic deep eutectic solvent: Synthesis, properties, and application in DNA separation

Summary: In this study, four hydrophobic magnetic deep eutectic solvents (HMDESs) were designed and prepared for DNA extraction, with the conformations of HMDESs simulated and their H-bonding interactions investigated using density functional theory (DFT) calculations. The physical and thermal properties of HMDESs were characterized, and DNA extracted by HMDESs could be quickly collected by an external magnet. Three auxiliary extraction methods were introduced, and factors impacting the DNA extraction process were systematically investigated through single-factor experimental analysis. The proposed extraction method successfully extracted DNA from complex matrices and E. coli cell lysate, and the binding sites between HMDESs and DNA were studied.

This study proposed a method to fabricate dual-layered Fe3O4@SiO2 nanoparticles with controlled SiO2 shell density and conducted structural, morphological, and magnetic analyses, revealing promising results for their application in biopurification. With the optimal conditions suggested based on the selective separation of plasmid DNA, this research contributes to the advancement of biomaterial purification techniques.
