DNA-Protein Cross-Linking Sequencing for Genome-Wide Mapping of Thymidine Glycol

Feng Tang et al.

Summary: The researchers developed a method called DPC-Seq for genome-wide mapping of Tg in human cells. By employing this method, they detected thousands of Tg sites and found that Tg is depleted in regions associated with active transcription but enriched at nucleosome-binding sites.

Click Chemistry Enables Rapid Amplification of Full-Length Reverse Transcripts for Long-Read Third Generation Sequencing

Eva S. Schoenegger et al.

Summary: Researchers developed a novel click chemistry-based method for generating and amplifying full-length cDNA libraries from total RNA, without the need for problematic template-switching reactions. This method enables amplification of previously inaccessible transcripts and provides larger insert sizes compared to the state-of-the-art click chemistry-based technique, PAC-seq.

Genome-wide analysis of 8-oxo-7,8-dihydro-2′-deoxyguanosine at single-nucleotide resolution unveils reduced occurrence of oxidative damage at G-quadruplex sites

Jiao An et al.

Summary: A method called CLAPS-seq was developed to accurately map the distribution of OGs in the genome. The study revealed decreased occurrence of OGs in G-quadruplexes and in promoters with high GC content. Additionally, potential quadruplex sequences were identified as hotspots of OGs, suggesting a role of non-G4 quadruplex sequences in the oxidative stress response mediated by OG.

Caught in motion: human NTHL1 undergoes interdomain rearrangement necessary for catalysis

Brittany L. Carroll et al.

Summary: Base excision repair (BER) is a key pathway in protecting cells from DNA damage caused by reactive oxygen species, with NTHL1 being a bifunctional DNA glycosylase that plays a role in repairing specific base damage; The X-ray crystal structure of hNTHL1 reveals an open conformation not seen in bacterial orthologs, with the flexibility and configuration change attributed to an interdomain linker; The large scale interdomain rearrangement during catalysis observed in hNTHL1 is unprecedented for a DNA glycosylase in the HhH superfamily, providing important insights into the molecular mechanism of this enzyme.
