Fine-tuning of aptamer complementary DNA for fluorescence resonance energy transfer assay

Summary: When developing a FRET method to detect target analytes, nucleic acid aptamers and complementary DNA (cDNA) play important roles. In this study, a ratio-based FRET analysis method was established for an OTA-oriented aptamer and its cDNAs using circular dichroism spectroscopy and molecular dynamic simulation. By optimizing the cDNA sequence, the fluorescence response ratio increased 13-fold and the detection limit was as low as 3.9 nM.
