Elevated sTREM2 and NFL levels in patients with sepsis associated encephalopathy

Gunseli Orhun et al.

Summary: The study investigates whether neurofilament light chain (NFL) and soluble triggering receptor expressed on myeloid cells 2 (sTREM2) could serve as prognostic biomarkers in sepsis-associated encephalopathy (SAE). The results show that serum and cerebrospinal fluid (CSF) levels of sTREM2 and NFL are associated with the severity of cognitive decline in SAE patients.

Neurofilament light chain levels predict encephalopathy and outcome in community-acquired pneumonia

Ha-Yeun Chung et al.

Summary: The objective of this study was to determine whether sNfL levels are increased in patients with community-acquired pneumonia (CAP) and if increased sNfL levels are associated with disease-associated confusion or disease severity. The study found that sNfL concentrations are specifically associated with confusion and not with pneumonia disease severity, thus reflecting a potential objective marker for encephalopathy in these patients.

Neurofilament light chain as neuronal injury marker - what is needed to facilitate implementation in clinical laboratory practice?

Burak Arslan et al.

Summary: Neurobiomarkers, particularly the neurofilament light chain protein (NfL), have received significant attention in the past decade. NfL has been developed as a widely used biomarker for axonal damage in various neurological disorders, including multiple sclerosis, amyotrophic lateral sclerosis, and Alzheimer's disease. Despite the availability of precise assays for NfL quantification, there are still analytical and logistical considerations that need to be addressed for its implementation in clinical practice. This review provides an overview of NfL as a biomarker and highlights the additional work needed to facilitate its clinical use.

Prognostic significance of SNCA and its methylation in bladder cancer

Zhengcun Wu et al.

Summary: This study investigates the role of SNCA gene in bladder cancer (BLCA) and its potential as a prognostic diagnostic biomarker. The results show that SNCA is downregulated in BLCA tumor tissues and is negatively correlated with DNA methylation. High SNCA expression is associated with poor overall survival. SNCA is also found to play a role in immune cell infiltration, particularly with T cells. Therefore, SNCA may serve as a promising prognostic biomarker in BLCA patients.

Summary: sNfL is a biomarker of neuronal damage used in monitoring disease activity in multiple sclerosis patients. This study established a reference database to correct for age and BMI, showing sNfL's potential in identifying individuals at risk for future disease activity. The results suggest that sNfL Z scores could predict acute and chronic disease activity in multiple sclerosis patients, with higher Z scores indicating increased risk.

Valerie J. Page et al.

Summary: This study aims to explore the association between plasma neurofilament light protein (NfL) and delirium in critically ill patients. The findings indicate that critically ill patients have high NfL levels upon admission, and patients with higher plasma NfL levels experience more delirium or deep sedation days. Additionally, measuring plasma NfL levels on the first day can predict mortality within six months.

Summary: This study aimed to assess the effects of demographics, lifestyle factors, and comorbidities on serum neurofilament light chain (sNfL) levels in individuals without neurologic disease and establish population-specific reference ranges for sNfL. The study analyzed data from the National Health and Nutrition Examination Survey (NHANES) and found that age, creatinine, and glycosylated hemoglobin (HbA1c) were the best predictors of sNfL levels. Injection of LPS from different bacterial strains in aged mice resulted in short-term neuroinflammation with subsequent normalization, showing more pronounced inflammatory responses with E. coli LPS compared to Salmonella LPS. Reduction in excitatory synaptic puncta in the hippocampus was observed at a later time point, possibly attributed to microglia-mediated elimination of C3-tagged synapses.
