Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Role of microbes in colorectal cancer therapy: Cross-talk between the microbiome and tumor microenvironment

Cong Xia et al.

Summary: The human gut microbiota plays a significant role in the development and progression of colorectal cancer, and manipulating the gut microbiota could be a novel approach for prevention and treatment. Specific bacteria have been found to have anti-tumor activity and can improve the tumor microenvironment by activating immune cells and upregulating the secretion of pro-tumor immune cytokines. This paper summarizes the potential benefits of certain bacteria in colorectal cancer, highlights their roles in the tumor microenvironment, discusses the application of gut microbes in combination with immunosuppressive agents, and provides suggestions for further experimental studies and clinical practice applications.

Somatic gene mutations expose cytoplasmic DNA to co-opt the cGAS/STING/NLRP3 axis in myelodysplastic syndromes

Amy F. McLemore et al.

Summary: This study reveals that NLRP3 inflammasome and IFN-stimulated gene (ISG) induction are key biological drivers of ineffective hematopoiesis and inflammation in myelodysplastic syndromes (MDS). Through the use of murine hematopoietic stem and progenitor cells and primary MDS BM specimens, it was found that accumulation of unresolved R-loops and micronuclei lead to the activation of the cytosolic sensor cyclic GMP-AMP synthase. Cyclic GMP-AMP synthase/stimulator of IFN genes (cGAS/STING) signaling causes ISG induction, NLRP3 inflammasome activation, and caspase-1 maturation.

Effect of miR-223-3p on cell pyroptosis in myelodysplastic syndrome and its mechanism via regulating the expression of NLRP3

Wanyi Yin et al.

Summary: This study aimed to investigate the regulatory mechanism of the miR-223-3p/NLRP3 signaling axis in the progression of myelodysplastic syndrome (MDS). The results showed that overexpression of miR-223-3p can effectively inhibit the expression of NLRP3 and cell pyroptosis in MDS.

Chronic stress promotes acute myeloid leukemia progression through HMGB1/NLRP3/IL-1β signaling pathway

Na Liu et al.

Summary: Chronic restraint stress promotes acute myeloid leukemia (AML) progression and mediates NLRP3 inflammasome activation in xenograft mice. HMGB1 regulates NLRP3 inflammasome activation in AML cells. Knockdown of HMGB1 inhibits AML progression under chronic stress in vivo.

Intracellular and Extracellular Lipopolysaccharide Signaling in Sepsis: Avenues for Novel Therapeutic Strategies

Ramy S. Gabarin et al.

Summary: Sepsis is characterized by organ dysfunction due to a dysregulated systemic host response to infection. Lipopolysaccharide (LPS) is a major mediator in sepsis, triggering inflammation through various pathways. Inhibiting intracellular LPS signaling by targeting caspases-4/5/1 or downstream effectors may be a promising therapeutic approach for sepsis treatment.

Therapeutic targeting of the inflammasome in myeloid malignancies

Samarpana Chakraborty et al.

Summary: Inflammatory microenvironment plays a critical role in modulating the development of myeloid malignancies by activating the innate immune system. Interactions between inflammasome mediators, immune modulators, and transcription factors are crucial in the pathogenesis of myeloid diseases and therapy resistance. Understanding the role of inflammasomes and immune pathways in myeloid malignancies, especially MDS/AML, is essential for developing effective therapeutic interventions.

NLRP3 Inflammasome Promotes the Progression of Acute Myeloid Leukemia via IL-1β Pathway

Chaoqing Zhong et al.

Summary: NLRP3 inflammasome is over-expressed and highly activated in AML bone marrow leukemia cells, promoting leukemia cell proliferation, inhibiting apoptosis and increasing resistance to chemotherapy. IL-1 beta plays a crucial role in this process. Targeting NLRP3 inflammasome may offer a novel therapeutic option for AML.

Impact of IL-1β and the IL-1R antagonist on relapse risk and survival in AML patients undergoing immunotherapy for remission maintenance

Hanna Grauers Wiktorin et al.

Summary: The study found that IL-1 beta serum levels were associated with survival rates in AML patients receiving immunotherapy for relapse prevention, with low IL-1 beta levels along with high IL-1RA levels being protective against leukemic relapse. Targeting IL-1 beta might impact on relapse risk and survival in AML.
