Single-cell sorting using integrated pneumatic valve droplet microfluidic chip

Summary: Single-cell analysis and sorting is an important technique in scientific research and industry. This study developed a microfluidic chip that integrates single-cell droplet generation, incubation, and sorting functions using a pneumatic micro-valve. Numerical simulations were used to optimize the design of the valve. Experimental results demonstrated that the integrated chip improved the purity of target droplets and reduced cell loss and labor. This design has various applications such as single-cell sequencing and antibody screening.

Multiple interactions between microfluidic droplets and on-chip pneumatic valves

Summary: In this study, the interaction between microfluidic droplets and pneumatic valves in droplet arrays was investigated experimentally and theoretically. Two main trapping modes were characterized, and the significant increase of hydrodynamic resistance induced by the droplet/valve interaction was quantified. Transitions between trapped and non-trapped regimes were rationalized using a generic theoretical model.

Summary: Extracellular vesicles (EVs) are cell-derived membrane structures that play a major role in physiological and pathological processes by delivering biomolecules to target cells. Many effects previously attributed to cell-based therapies are actually mediated by EVs, suggesting the use of purified EVs instead of living cells for therapy. This review focuses on the heterogeneity of EVs derived from mesenchymal stem/stromal cells (MSC) and summarizes the upstream parameters that critically affect their therapeutic properties and biological functions.

Investigation of daughter cell dissection coincidence of single budding yeast cells immobilized in microfluidic traps

Summary: Microfluidic methodologies for replicative lifespan determination of budding yeast cells are highly impacted by the robustness of experimental conditions, especially the design of yeast-trapping structures. In this study, an optimized design, the yeast rotation (YRot) trap, was proposed and validated for stable retention of mother cells and hydrodynamic rotation of buds. The integration of denser YRot traps in microchannels could potentially improve experimental conditions for precise RLS determination in yeast aging studies.

Heterogeneity in Human Induced Pluripotent Stem Cell-derived Alveolar Epithelial Type II Cells Revealed with ABCA3/SFTPC Reporters

Summary: In this study, a new ABCA3:GFP reporter was generated for the purification of induced pluripotent stem cell-derived alveolar epithelial type 2 cells (iAEC2s) and visualization of organelles associated with AEC2 maturation. Comparative RNA sequencing and functional studies revealed cellular heterogeneity within the iAEC2 population, with ABCA3:GFP single-positive cells exhibiting faster growth kinetics and higher pluripotency compared to ABCA3:GFP/SFTPCtdTomato double-positive cells, which expressed more maturation markers.
