Real-time visualization of lysosomal pH fluctuations in living cells with a ratiometric fluorescent probe

Summary: A lysosome-targeted fluorescent probe Lyso-CMCP was developed for real-time monitoring pH changes in living cells. This probe showed sensitive and selective response to acidic pH and had favorable features including good photostability, reversibility, anti-interference ability, and low cytotoxicity. The lysosomal pH fluctuations were successfully monitored and quantified by ratiometric fluorescence imaging using this probe.

Imidazolyl-benzocoumarins as ratiometric fluorescence probes for biologically extreme acidity

The probe, a push-full type 3-(imidazolyl)benzocoumarin dye, has the lowest pK(a) value among known ratiometric probes. The probe has high quantum yields, chemical stability and aqueous solubility, and it has been successfully used for ratiometric fluorescence imaging of intrabacterial acidity, providing a practical means for studying biological systems under extreme pH conditions.
