Study on Asymmetric Conjugate Cyanation of 3,5-Dimethyl-N-α,β-unsaturated Acylpyrazole Catalyzed by Magnesium-Bisoxazoline Complex

Liu Yunshu et al.

Summary: The asymmetric conjugate cyanation reaction of TMSCN with 3,5-dimethyl-N-alpha,beta-unsaturated acylpyrazole catalyzed by magnesium-bisoxazoline complex was studied. The results showed that by generating chiral catalyst under specific conditions, high yields and enantioselectivities could be achieved.

A synthetic approach to 5/5/6-polycyclic tetramate macrolactams of the discodermide type

Kevin Bodenschatz et al.

Summary: A flexible synthetic route to the 16-membered tetramate-embedding macrocyclic scaffold present in various polycyclic tetramate macrolactams (PTMs) was developed. The approach differs from previous studies in terms of protecting groups and the order of connections. Additionally, a short approach to various stereoisomers of the 5/5/6-tricarbocyclic motif was devised starting from Weiss' diketone.

Review of Voltage-gated Calcium Channel α2δ Subunit Ligands for the Treatment of Chronic Neuropathic Pain and Insight into Structure-activity Relationship (SAR) by Pharmacophore Modeling

Yuting Chen et al.

Summary: This study reviewed the latest progress in the discovery and development of gabapentinoids for the treatment of chronic neuropathic pain (NP). The structure-activity relationship (SAR) of gabapentinoids was analyzed through pharmacophore modeling. The findings provide valuable insights for the future design of novel therapeutic agents targeting the voltage-gated calcium channel (VGCC) alpha 2 delta subunit.

NEUROPATHIC PAIN: FROM MECHANISMS TO TREATMENT

Nanna Brix Finnerup et al.

Summary: Neuropathic pain is a common chronic pain condition caused by lesion or disease of the somatosensory nervous system, characterized by ongoing or intermittent spontaneous pain. Evoked pain may spread to neighboring areas and is associated with peripheral and central sensitization processes.

Pathogenesis, diagnosis and clinical management of diabetic sensorimotor peripheral neuropathy

Summary: This article discusses the latest advances in the mechanisms of DSPN and painful DSPN, as well as the emerging diagnostics and treatments. While our understanding of the disease has come from animal models, effective therapies in preclinical models have not translated into clinical trials. Management of painful DSPN remains suboptimal, but a mechanism-based approach may offer enhanced pain relief.
