Development of an efficient wastewater testing protocol for high-throughput country-wide SARS-CoV-2 monitoring

Diyar Mailepessov et al.

Summary: Wastewater-based surveillance is a widely used non-intrusive tool for monitoring population-level transmission of COVID-19. This study aimed to identify and evaluate SARS-CoV-2 virus concentration protocols for high-throughput wastewater testing. The results showed that optimal virus recovery was achieved by removing debris, incubating with 8% PEG at 4 degrees C, centrifugation, and column-based RNA extraction methods.

Comparison of RT-qPCR and RT-dPCR Platforms for the Trace Detection of SARS-CoV-2 RNA in Wastewater

Warish Ahmed et al.

Summary: This study compared the performance of RT-qPCR and RT-dPCR platforms in detecting SARS-CoV-2 RNA in low-prevalence locations in Queensland, Australia. The results showed that RT-dPCR had a lower limit of detection and a higher detection rate than RT-qPCR in both eluate and pellet samples.

Pan-Enterovirus Amplicon-Based High-Throughput Sequencing Detects the Complete Capsid of a EVA71 Genotype C1 Variant via Wastewater-Based Epidemiology in Arizona

Temitope O. C. Faleye et al.

Summary: A genotype C1-like Enterovirus A71 variant was found in wastewater in a neighborhood in Tempe, Arizona, circulating for over two years without its sequence being documented. The study emphasizes the importance of wastewater-based epidemiology (WBE) as an early warning system for virus surveillance, especially in light of changes caused by the COVID-19 pandemic.

A comparison of SARS-CoV-2 wastewater concentration methods for environmental surveillance

Sarah E. Philo et al.

Summary: Wastewater surveillance of SARS-CoV-2 can be a helpful complement to clinical surveillance due to asymptomatic cases and potential testing limitations. Different methods were evaluated for concentration and recovery of SARS-CoV-2 and OC43, with skimmed milk flocculation showing the best results and feasibility in low-resource settings.

Comparing analytical methods to detect SARS-CoV-2 in wastewater

Alba Perez-Cataluna et al.

Summary: Wastewater based epidemiology is a reliable strategy for assessing the COVID-19 pandemic, with recent studies showing the feasibility of detecting SARS-CoV-2 in wastewater. Different methods for concentration and extraction were evaluated, with the aluminum precipitation method coupled with automated nucleic extraction proving to be a sensitive and acceptable method for surveillance of SARS-CoV-2 in wastewater.

Evaluation of viral concentration methods for SARS-CoV-2 recovery from wastewaters

Patricia Angelica Barril et al.

Summary: Wastewater-based epidemiology is a valuable tool for monitoring SARS-CoV-2 in communities and early detection of COVID-19 outbreaks. This study evaluated 11 different virus concentration techniques in wastewater and found that methods involving PEG precipitation and PAC flocculation showed high efficiency for virus recovery. Further testing showed that PAC flocculation had a lower limit of detection for SARS-CoV-2 compared to PEG precipitation.

Development of a large volume concentration method for recovery of coronavirus from wastewater

Brian R. McMinn et al.

Summary: Levels of SARS CoV 2 RNA in wastewater can be used to monitor COVID-19 in communities, but current detection methods have limitations in processing sufficient volumes of source material. This study evaluated a two-stage concentration method for recovering OC43 virus from wastewater, optimizing efficiency for potential use in wastewater surveillance.

Reproducibility and sensitivity of 36 methods to quantify the SARS-CoV-2 genetic signal in raw wastewater: findings from an interlaboratory methods evaluation in the US

Brian M. Pecson et al.

Summary: In response to COVID-19, the international water community developed methods to quantify the SARS-CoV-2 genetic signal in untreated wastewater. An interlaboratory assessment evaluated the reproducibility and sensitivity of 36 standard operating procedures (SOPs), showing that a variety of methods are capable of producing reproducible results and emphasizing the importance of selecting the same SOP or laboratory to track SARS-CoV-2 trends.
