Computational assessment of the impact of Cu(II) and Al(III) on β-amyloid42 fibrils: Binding sites, structural stability, and possible physiological implications

Summary: One of the major features of Alzheimer's disease is the aggregation of beta-amyloid peptide, which is influenced by metal ions. This study used computational methods to investigate the binding regions and conformational effects of Cu(II) and Al(III) ions on the fibrillary structure of beta-amyloid (A beta(42)). The results showed that Cu(II) binding disrupted the structure, while Al(III) binding enhanced stability. These findings provide insights into the molecular impact of metal ion interactions in Alzheimer's disease progression.

Summary: Alzheimer's disease (AD) is a common form of dementia and its causes are not fully understood. High concentration of certain metal cations has been linked to the development of AD and the production of reactive oxygen species that cause neuronal damage. Multifunctional ligands have been proposed as a potential treatment to prevent or reverse the damaging effects of these metal cations. This review discusses recent advances in understanding the role of metal cations and the development of multifunctional ligands for AD.

Mechanism by Which Aluminum Regulates the Abnormal Phosphorylation of the Tau Protein in Different Cell Lines
