They are involved not only in maintaining metabolic and ionic balance, but also in cognitive functions and information processing. Controversies exist regarding their role in aging and age-related neurodegenerative diseases, such as Alzheimer's disease. Recent research has focused on neuron-glial interactions and the role of astrocytes in sleep physiology and neurodegenerative diseases, as well as the reorganization of astrocytic networks during aging.

Neuronally Derived Soluble Abeta Evokes Cell-Wide Astrocytic Calcium Dysregulation in Absence of Amyloid Plaques in Vivo

Summary: The key pathologic entities driving the destruction of synaptic function and integrity during the evolution of Alzheimer's disease (AD) remain elusive. Astrocytes, through calcium-based physiology, play a central role in AD pathogenesis by modulating synaptic transmission, vascular dynamics, and neurovascular coupling. In this study, we found that the calcium signaling within capillary endfeet of awake APP/PS1 transgenic mice was reduced compared to astrocytic fine processes. Additionally, the presence of amyloid plaques led to an increase in intracellular calcium in cortical astrocytes and dys-regulation of spontaneous calcium signaling.

Capillary dysfunction correlates with cortical amyloid load in early Alzheimer's disease

Summary: Alterations in cerebral perfusion play a crucial role in Alzheimer's disease (AD). Deficiencies in brain microvascular circulation, together with accumulated amyloid-β, may result in local hypoxia. This study investigated alterations in cerebral circulation and the correlation with amyloid-β load in prodromal AD (pAD). The results showed that the pAD-MCI group had significantly lower microvascular blood flow and higher heterogeneity of microvascular blood transit times compared to other groups. Additionally, elevated amyloid-β levels correlated with decreased microvascular blood flow and increased heterogeneity in frontal and temporal areas of the pAD-MCI group.

d-Galactose and Hypoxia Induce the Early Onset of Age-Related Hearing Loss Deterioration in a Mouse Model

Summary: This study found that aging accelerates after a long period of exposure to hypoxia and environmental change, but not genetic modifications. By injecting d-galactose and exposing the mice to hypoxia, early-onset age-related hearing loss was induced within a short time. This research suggests that environmental stimulation can induce age-related hearing loss in a murine model.

Chronic hypoxia of endothelial cells boosts HIF-1α-NLRP1 circuit in Alzheimer's disease

Summary: The cerebral microvasculature of Alzheimer's disease (AD) patients exhibits reduced capillary diameter and impaired blood flow. This study found that the AD mouse model showed hypoxic vessels expressing hypoxia-inducible factor-1 alpha (HIF-1 alpha) and NLRP1 inflammasome formation. In vitro experiments demonstrated that hypoxic endothelial cells expressed NLRP1 and showed upregulation of ASC-caspase-1-IL-1 beta cascades. These findings suggest that chronic hypoxia in microvascular endothelial cells can trigger inflammation and further damage the vascular system in AD.

Novel Algorithm of Network Calcium Dynamics Analysis for Studying the Role of Astrocytes in Neuronal Activity in Alzheimer's Disease Models

Summary: This study investigates the impact of astrocytes on the calcium activity of neuron-astroglia networks in Alzheimer's disease (AD) modeling. The researchers developed an algorithm to analyze the dynamic directed graphs of both astrocytic and neuronal networks, allowing them to identify functional relationships and characterize the spread of calcium signal. The study found that Alzheimer's astrocytes can alter the functional pattern of calcium activity in healthy nerve cells.

Astrocyte dystrophy in ageing brain parallels impaired synaptic plasticity

Summary: This study found a significant reduction in the number and length of astrocytic processes, astrocytic territorial domains, and astrocyte-to-astrocyte coupling in the aged brain. Physiological deficits in K+ and glutamate clearance, as well as spatiotemporal reorganisation of Ca2+ events, were observed in old astrocytes, which paralleled impaired synaptic long-term potentiation (LTP) in hippocampal CA1 in old mice. These findings may explain the astroglial mechanisms of age-dependent decline in learning and memory.

One-Year Prospective Study of Plasma Biomarkers From CNS in Patients With Mild Traumatic Brain Injury

Summary: The study showed that GFAP and tau levels were highest in the acute phase, while NFL peaked at 2 weeks and 3 months in patients with mTBI. Patients with concomitant other somatic injuries had the most significant elevations in biomarker levels, and were more likely to be victims of traffic accidents and violence. Acute GFAP concentrations displayed the highest discriminability between patients and controls.
