Protein nanopore reveals the renin-angiotensin system crosstalk with single-amino-acid resolution

Summary: A nanopore framework is used to quantitatively monitor the crosstalk effects between angiotensin-converting enzyme (ACE) and angiotensin-converting enzyme 2 (ACE2) on the renin-angiotensin system (RAS). ACE2 selectively inhibits ACE to prevent cleavage of angiotensin I, even at a higher concentration of ACE. The spike protein of SARS-CoV-2 suppresses the activity of ACE2, leading to the accumulation of angiotensin II and relaxation of ACE. The Delta variant has a greater impact on the crosstalk than the wild type.

A novel specific and ultrasensitive method detecting extracellular vesicles secreted from lung cancer by padlock probe-based exponential rolling circle amplification

Summary: A highly specific and sensitive fluorescence scheme based on padlock probe-based exponential rolling circle amplification (P-ERCA) assay for detecting extracellular vesicles derived from lung cancer cells was reported. The method exhibits high specificity and sensitivity, and successfully distinguishes between lung cancer patients and healthy individuals in real human serum samples.

Bioinspired Hydrogen Peroxide-Activated Nanochannels and Their Applications in Cancer Cell Analysis

Summary: In this study, artificial nanochannels activated by hydrogen peroxide (H2O2) were designed and constructed, showing a highly selective and sensitive response to H2O2. The mechanism of H2O2-triggered regulation of ion transport by the nanochannels was evidenced through ionic current tests and comsol simulations. The functionalized nanochannel membrane was applied for the detection of H2O2 in the tumor microenvironment, achieving highly selective distinguishing of cancerous cells from normal cells.

Integrating Ordered Two-Dimensional Covalent Organic Frameworks to Solid-State Nanofluidic Channels for Ultrafast and Sensitive Detection of Mercury

Summary: This study demonstrates the integration of ordered two-dimensional covalent organic frameworks (2D COFs) into solid-state nanofluidic channels for rapid, selective, and sensitive detection of contaminants. The JNU-3@AAO-based nanofluidic sensor allows for ultrafast and precise determination of Hg(II), with a low limit of detection, wide linear range, and good precision. The developed method is also successfully applied to the determination of mercury in complex samples.

Ionic Signal Enhancement by the Space Charge Effect through the DNA Rolling Circle Amplification on the Outer Surface of Nanochannels

Peptide Nucleic Acid-Functionalized Nanochannel Biosensor for the Highly Sensitive Detection of Tumor Exosomal MicroRNA

Summary: A PNA-functionalized nanochannel biosensor was proposed for the ultrasensitive and specific detection of tumor exosomal miRNAs, achieving a detection limit as low as 75 aM. The biosensor could effectively distinguish pancreatic cancer tumor-derived exosomes from the normal control group, and the results were consistent with qRT-PCR method. Moreover, the biosensor showed potential for early cancer diagnosis by detecting exosomal miRNA-10b in clinical plasma samples with higher content in cancer patients compared to healthy individuals.

Revealing Ionic Signal Enhancement with Probe Grafting Density on the Outer Surface of Nanochannels

Summary: This study investigates the impact of probe grafting density on the outer surface of nanochannels on the change rate of ionic signals before and after target recognition. The experimental results show that with the same number of targets, the beta value increases with the probe grafting density. A theoretical model is established with clearly defined physical properties of each probe and target, while numerical simulations suggest that background current decrease and target aggregation contribute to signal enhancement. This work reveals the signal mechanism of probe-target recognition and suggests a general approach to sensitive nanochannel/nanopore design with good selectivity.
