Identification of damage associated molecular patterns and extracellular matrix proteins as major constituents of the surface proteome of lung implantable silicone/nitinol devices

Akash Gupta et al.

Summary: This study investigates the composition of the adhered proteome on explanted lung devices, which are widely used as mechanical interventions for pulmonary pathologies. The research reveals that long-term efficacy can be impacted by fibrotic or granulation tissue formation around the implant sites. The study identifies 263 unique protein species that are mutually adsorbed on the explanted lung devices, and pathway analyses suggest their association with coagulation, pattern recognition receptor signaling, immune responses, cytoskeleton organization, cell adhesion, and migration. The findings also highlight the importance of extracellular matrix proteins and damage-associated molecular patterns in the formation of the surface proteome.

Quantum Sensing of Free Radicals in Primary Human Dendritic Cells

Linyan Nie et al.

Summary: Free radicals are important indicators for stress and pathogenic conditions, and are difficult to detect due to their reactivity and low abundance. A novel relaxometry method using a fluorescent defect in a diamond has been developed for subcellular detection of radicals with nanoscale MRI capabilities. Variability in radical production was observed in primary dendritic cells from healthy donors, and their response to stimulation or inhibition was investigated.

STAT1 and Its Crucial Role in the Control of Viral Infections

Manlio Tolomeo et al.

Summary: STAT1 protein plays a crucial role in the immune response against viruses, and its deficiency can result in severe infections and even death. It has been found that some viruses inhibit the activity of STAT1, making it important to search for drugs that can target these viral proteins for the treatment of dangerous viral infections and future viral pandemics, as demonstrated by the clinical results with Paxlovid in SARS-CoV-2-infected patients.

Titanium dioxide nanoparticles induced reactive oxygen species (ROS) related changes of metabolomics signatures in human normal bronchial epithelial (BEAS-2B) cells

Jiahe Zhang et al.

Summary: This study explored the impact of titanium dioxide nanoparticles (TiO2 NPs) on metabolomics signatures in human normal bronchial epithelial cells and investigated the role of reactive oxygen species (ROS). The results showed that TiO2 NPs significantly altered the lipid metabolism and increased intracellular ROS levels, indicating an oxidative stress response.

JAK/STAT Signaling: Molecular Targets, Therapeutic Opportunities, and Limitations of Targeted Inhibitions in Solid Malignancies

Bilal Rah et al.

Summary: The JAK/STAT signaling pathway plays an important role in regulating cellular processes such as immune regulation, cell proliferation, survival, apoptosis, and hematopoiesis. Dysregulation of this pathway is associated with various immunological disorders, hematological diseases, and solid malignancies. Understanding the intricate mechanisms of JAK/STAT signaling in solid malignancies is crucial for drug development and improving cancer therapies.

Applying NV center-based quantum sensing to study intracellular free radical response upon viral infections

Kaiqi Wu et al.

Summary: This study demonstrates the utility of diamond magnetometry in studying the free radical response during Semliki Forest virus infection. The alterations in free radical concentration near infectious viruses were measured using the spin-lattice relaxation of NV defect ensembles in intracellular nanodiamonds. The study provides insights into the real-time free radical variation during the early infectious process.

Effect of Nitinol surface with nanotubes and/or ordered nanopores on cell behavior

Katerina Vrchovecka et al.

Summary: Recent medical applications have specific requirements on materials, and Nitinol can fulfill them by modifying its surface characteristics. Various nanostructuring methods can enhance the oxide layer on the Nitiniol surface, resulting in improved biocompatibility and corrosion resistance. Certain dimensions of nanotubes have a positive effect on the adhesion and viability of osteoblasts and endothelial cells, while having a negative effect on smooth muscle cells. Also, the topography of nanostructures and the release rates of Ni ions are correlated with increased antibacterial effect.

Airway granulation response to lung-implantable medical devices: a concise overview

Sharyn A. Roodenburg et al.

Summary: Long-term effectiveness and sustainability of airway implantable devices are hindered by the reaction of granulation tissue formation, but research on underlying mechanisms and risk factors is scarce. Factors such as procedural-related tissue injury, micro-organism presence, device design, and patient-related factors all play a role in the severity of tissue response and granulation tissue formation. Collaboration between scientific communities is needed to improve understanding and enhance the long-term treatment effectiveness of lung-implantable devices.
