Menopausal hormone therapy and risk of biliary tract cancers

Sarah S. Jackson et al.

Summary: A nested case-control study found that the use of menopausal hormone therapy (MHT) may be associated with the risk of gallbladder cancer (GBC) and cholangiocarcinoma (CCA), but not ampulla of Vater (AVC) or mixed cancer. The specific formulation and administration of MHT have different effects on the risk.

Clonally expanded B cells in multiple sclerosis bind EBV EBNA1 and GlialCAM

Tobias Lanz et al.

Summary: Multiple sclerosis (MS) is an autoimmune disease where autoreactive lymphocytes attack the central nervous system. Epstein-Barr virus (EBV) infection has been linked to MS, with molecular mimicry between EBV nuclear antigen 1 (EBNA1) and central nervous system protein GlialCAM. EBNA1 immunization exacerbates disease in MS mouse models and anti-EBNA1 and anti-GlialCAM antibodies are prevalent in MS patients, providing a mechanistic link between MS and EBV and guiding the development of new MS therapies.

Beneficial autoimmunity and maladaptive inflammation shape epidemiological links between cancer and immune-inflammatory diseases

Jonathan Pol et al.

Summary: Chronic inflammation increases the risk of malignant transformation through proliferative responses. Autoimmune responses can both drive chronic inflammation and enhance cancer immunosurveillance. Recent study confirmed the negative associations between breast cancer and rheumatoid arthritis or systemic lupus erythematosus.

Potential Role of Inflammation-Promoting Biliary Microbiome in Primary Sclerosing Cholangitis and Cholangiocarcinoma

Katsuyuki Miyabe et al.

Summary: This study investigated the microbiota profiles in bile and stool samples of patients with PSC and CCA, and found that bile and stool have different microbiota profiles. The abundance of Fusobacteria in bile was correlated with the duration of PSC in CCA patients. These findings suggest a potential role for microbiota-driven inflammation in the pathogenesis of perihilar CCA.

Hepatocellular Carcinoma in Primary Biliary Cholangitis

Alexander M. Sy et al.

Summary: Several studies have shown that patients with PBC have an increased risk of developing HCC. Risk factors for HCC in PBC include advanced age, male sex, comorbidities, and advanced liver fibrosis. The role of treatment response as a risk factor for HCC development in PBC patients should be further investigated. HCC in PBC has a poor prognosis compared to other chronic liver diseases with HCC, and liver transplantation offers the highest survival rate among available therapies.

Incidence of Hepatocellular Carcinoma in Primary Biliary Cholangitis: A Systematic Review and Meta-Analysis

Summary: The risk factors for hepatocellular carcinoma (HCC) in patients with primary biliary cholangitis (PBC) are not clear, but cirrhosis and male gender are shown to be significant factors in increasing HCC incidence. Further studies are needed to evaluate additional risk factors in this population and support current recommendations for HCC surveillance in PBC patients with cirrhosis.

Decreased infiltration of CD4+ Th1 cells indicates a good response to ursodeoxycholic acid (UDCA) in primary biliary cholangitis

Kangjie Yu et al.

Summary: The infiltration of CD4+ T cells in PBC patients is significantly associated with the response to UDCA treatment, with a decrease in infiltration leading to a better response. However, changes in CD3+ T cells, CD8+ T cells, and CD20+ B cells do not show significant differences between responders and nonresponders. Additionally, the number of T-bet+ Th1 cells in the liver after UDCA treatment is significantly different between responders and nonresponders, indicating its potential use in evaluating therapeutic response.

Gut-Liver Immune Traffic: Deciphering Immune-Pathogenesis to Underpin Translational Therapy

Amber G. Bozward et al.

Summary: The close relationship between the gastrointestinal tract and liver constitutes the gut-liver axis, playing a central role in the pathogenesis of gut-liver axis diseases. This axis is crucial for immune homeostasis, influenced by factors such as the gut microbiota, diet, genetics, and environment. Through the coordinated effort of immune cells network, the liver is able to maintain its tolerogenic state.
