Direct Cytosolic Delivery of Citraconylated Proteins

Summary: Current strategies for intracellular protein delivery face challenges of endosomal entrapment and degradation. This study presents a simple approach of protein modification using citraconic anhydride to impart an overall negative charge, enabling assembly with positively charged nano vectors. Anhydride-modified proteins were electrostatically assembled with a cationic guanidinium-functionalized polymer, demonstrating efficient cytosolic protein delivery. The platform retained protein function and effectively killed cells through intracellular enzymatic activity.

Inorganic nanoparticles as scaffolds for bioorthogonal catalysts

Bioorthogonal nanozymes for breast cancer imaging and therapy

Summary: Bioorthogonal catalysis via transition metal catalysts (TMCs) allows the production of therapeutics locally through chemical reactions that are inaccessible to biological systems. This localized approach can improve the effectiveness of anticancer treatment and minimize off-target effects. By encapsulating TMCs into nanomaterials, nanozymes are created to activate imaging and therapeutic agents. In this study, cationic bioorthogonal nanozymes were used to establish localized drug factories for cancer therapy in vivo. These nanozymes remained at the tumor site for at least seven days after a single injection due to interactions with cell membranes and tissue components. The non-toxic prodrugs were then systemically administered and continuously converted into active drugs by the nanozymes. This strategy significantly reduced tumor growth in an aggressive breast cancer model, with reduced liver damage compared to traditional chemotherapy.

A computational workflow written in Python is described to achieve accurate registration between datasets from these two imaging modalities, allowing for pixel-by-pixel comparisons. The registration process provides deeper insights into how nanomaterial delivery systems influence lipid biochemistry in tissues and allows for improved segmentation in MALDI-MS images for identification of lipids associated with different sub-organ regions in tissues.
