MicroRNA-200c-targeted contactin 1 facilitates the replication of influenza A virus by accelerating the degradation of MAVS

Shuai Xu et al.

Summary: In this study, the author identified CNTN1 as a critical host factor for the replication of IAV and found that it promotes viral replication by negatively regulating the RIG-I-mediated type I interferon pathway. Mechanistically, CNTN1 enhances the proteasomal degradation of MAVS by promoting USP25-mediated deubiquitination. These findings uncover novel roles of the host protein CNTN1 in type I interferon signaling and IAV replication.

Ordered assembly of the cytosolic RNA-sensing MDA5-MAVS signaling complex via binding to unanchored K63-linked poly-ubiquitin chains

Bin Song et al.

Summary: The study reveals that long unanchored K63-polyUb chains (n ≥ 8) mediate tetramerization of MDA5CARDs, bridging MDA5CARDs and MAVS into proximity. ATP binding and hydrolysis stabilize RNA-bound MDA5 and promote MAVS activation via allosteric effects on CARDs-polyUb complex.

USP18 positively regulates innate antiviral immunity by promoting K63-linked polyubiquitination of MAVS

Jinxiu Hou et al.

Summary: Research demonstrates that deubiquitinase USP18 regulates MAVS-mediated antiviral signaling by promoting MAVS interaction with TRIM31, leading to increased production of type I interferon and enhanced resistance to RNA viruses.
