Pivotal role of the protein corona in the cell uptake of fluorinated nanoparticles with increased sensitivity for F-19-MR imaging

Water-Soluble Chemically Precise Fluorinated Molecular Clusters for Interference-Free Multiplex 19F MRI in Living Mice

Summary: This study presents a water-soluble molecular 19F MRI nanoprobe for multispectral color-coded 19F MRI by conjugating fluorine-containing moieties with a polyhedral oligomeric silsesquioxane (POSS) cluster. The chemically precise fluorinated molecular clusters exhibit excellent aqueous solubility, high 19F contents, and suitable relaxation times for high-performance 19F MRI. Three POSS-based molecular nanoprobes with distinct 19F chemical shifts were constructed, enabling interference-free multispectral color-coded 19F MRI of labeled cells in vitro and in vivo. In vivo experiments demonstrated selective accumulation of these molecular nanoprobes in tumors and subsequent rapid renal clearance, highlighting their promising in vivo behavior for biomedical applications. This study provides an efficient strategy to expand the 19F probe libraries for multispectral 19F MRI in biomedical research.

Optimization of superfluorinated PLGA nanoparticles for enhanced cell labelling and detection by 19F-MRI

Summary: Fluorine-19 (19F) magnetic resonance imaging (MRI) is an emerging technique for molecular imaging and cell tracking. This paper reports the development of a superfluorinated tracer called PERFECTA, as well as the use of differently coated PLGA NPs as promising 19F-MRI probes. The results show that the stabilized NPs with anionic surfactant exhibit optimal cellular uptake and shorter T1 relaxation times, enhancing the sensitivity of 19F-MRI.

Correlating Corona Composition and Cell Uptake to Identify Proteins Affecting Nanoparticle Entry into Endothelial Cells

Summary: The formation of a biomolecule corona on nanoparticles affects their performance in drug delivery. By analyzing correlations between the corona composition of silica nanoparticles and their uptake by endothelial cells, proteins influencing nanoparticle uptake were identified. Precoating nanoparticles with histidine-rich glycoprotein decreased uptake in brain and liver endothelium, while the transferrin receptor was found to play a role in nanoparticle uptake in liver endothelium. These findings suggest that the composition of the protein corona can be manipulated to alter nanoparticle uptake by cells.
