P2X7 receptor involved in antitumor activity of atractylenolide I in human cervical cancer cells

Yue Han et al.

Summary: This study found that Atr-I has antitumor activity in human cervical cancer cells and P2X7R is involved in this effect. The combination of Atr-I and P2X7R agonist BzATP can restore the inhibitory effect of Atr-I in Hela cells, while the combination of Atr-I and P2X7R antagonist JNJ has an additive effect on cell growth inhibition in SiHa cells.

P2X7 receptor: the regulator of glioma tumor development and survival

Damian Matysniak et al.

Summary: P2X7 receptor, known for its ionotropic nucleotide properties, has a wide range of effects on glioma biology, including promoting cell proliferation, enhancing cell adhesion, and influencing mitochondria depolarization and reactive oxygen species production. In vivo studies show that P2X7 activation in glioma tumors leads to tumor growth via pro-survival signaling pathways, but treatment with a selective antagonist inhibits tumor development and modulates the immune response. Upregulation of P2X7 receptor is also observed in human gliomas grades I to III, indicating its potential role in the disease progression.

Detection and Functional Evaluation of the P2X7 Receptor in hiPSC Derived Neurons and Microglia-Like Cells

Linda Francistiova et al.

Summary: Experimental results show the expression of P2X7 receptors on human hiPSC-derived cells, including microglia-like cells and neuronal cells. However, the receptors are not localized on the cell membrane in neuronal cells, suggesting they may not directly mediate neurotoxicity. Additionally, there are no functional differences in P2X7 receptors between control and familial Alzheimer's disease patient-derived neuronal cells.

Integrating nonlinear analysis and machine learning for human induced pluripotent stem cell-based drug cardiotoxicity testing

Andrew Kowalczewski et al.

Summary: Using hiPSC technology, nonlinear analysis, and machine learning, we can create novel tools to assess drug-induced cardiotoxicity on human cardiomyocytes. By extracting parameters, applying dimensionality reduction techniques, and incorporating nonlinear analysis, we improve the classification accuracy of drug effects on cardiac contractions and predict cardiotoxicity levels.

Ketamine impairs growth cone and synaptogenesis in human GABAergic projection neurons via GSK-3β and HDAC6 signaling

Xuan Li et al.

Summary: This study revealed the mechanisms by which ketamine affects the growth cones and synapse formation of developing GABAergic neurons using human pluripotent stem cells. The results indicate that ketamine impairs growth cone formation, synaptogenesis, and dendrite maturation through the activation of GSK-3β pathway and inhibition of HDAC6.

A fully-automated paper ECG digitisation algorithm using deep learning

Huiyi Wu et al.

Summary: There has been increasing attention on using deep learning methods to analyze electrocardiograms (ECGs), as recent studies have shown that neural networks (NNs) can predict heart failure or atrial fibrillation based solely on ECG signals. However, many ECGs are only available in paper format, which is not suitable for training NNs. In this study, a fully-automated online ECG digitization tool was developed to convert scanned paper ECGs into digital signals. The algorithm successfully segmented the ECG image into separate images for each lead and achieved high correlation between the digitized signals and the ground truth ECG.

Compact automated culture machine for human induced pluripotent stem cell maintenance and differentiation

Kazunori Bando et al.

Summary: This study developed an all-in-one automated culture machine that can efficiently expand human pluripotent stem cells and differentiate them into various cell types. This would facilitate the social implementation of human pluripotent stem cells and contribute to the standardization of the entire handling processes.

Single-cell Stereo-seq reveals induced progenitor cells involved in axolotl brain regeneration

Xiaoyu Wei et al.

Summary: This study used Stereo-seq technology to analyze spatial distribution and molecular features of axolotl brain during development and regeneration. The researchers identified an injury-induced ependymoglial cell cluster as a potential progenitor cell population for replenishing lost neurons. They also discovered regressed neurons and provided evidence for the involvement of resident ependymoglial cells in neurogenesis.

Summary: We present a high-content analysis protocol for monitoring the outgrowth capacity of human neurons derived from iPSCs. This protocol involves imaging and quantifying the morphology of dendrites and axons within a high-throughput system. It can be used to screen for modulators of neuronal morphogenesis or neurotoxicity and applied to patient-derived iPSCs.
