Summary: Two new diterpenoids, penicichrysogene A and penicichrysogene B, were isolated from the endophytic fungus Penicillium chrysogenum MT-12, which was obtained from the medicinal plant Huperzia serrata. Their structures were determined based on extensive spectroscopic and spectrometric data. Compound 1 exhibited inhibitory activity on ATP release from thrombin-activated platelets.

Efficient exploration of terpenoid biosynthetic gene clusters in filamentous fungi

Summary: In this study, an automated and high-throughput bioengineering workflow using Aspergillus oryzae as a chassis was developed, which enables efficient genome mining, characterization of biosynthetic gene clusters (BGCs), and identification of bioactive fungal terpenoids. By simultaneously recombining 39 BGCs, a total of 185 distinct terpenoids were produced, and one of them exhibited anti-inflammatory properties. Furthermore, the mevalonate pathway in A. oryzae was optimized to enhance terpenoid production.

Discovery, Structure, and Mechanism of a Class II Sesquiterpene Cyclase

Summary: This study reports the gene mining and structural characterization of two class II sesquiterpene cyclases from Streptomyces. The high-resolution X-ray crystal structures of these cyclases unveiled an induced-fit mechanism with a unique Mg2+ binding mode, solving one of the lingering questions in class II TC enzymology. This study provides new insights into class II TCs and supports further genome mining and TC engineering.

Discovery of branching meroterpenoid biosynthetic pathways in Aspergillus insuetus: involvement of two terpene cyclases with distinct cyclization modes

Summary: A new meroterpenoid biosynthetic gene cluster was discovered in Aspergillus insuetus CBS 107.25, where a terpene cyclase folds the precursor compound 1 into a new meroterpenoid species with a different conformation. Another cyclase in the strain also synthesizes a distinct cyclized product of 1, resulting in diverging of the biosynthetic pathway in the fungus.

Summary: The study analyzed the volatile oil produced through steam distillation of lavender to establish the essential oil yield and aromatic profile from different parts of the plant. The essential oils from the corolla, calyx, leaf, and whole flowering top were analyzed and found to contain similar compounds but in varying relative percentages. The aromatic profiles of the different plant parts provide insight into the influence of the corolla, calyx, and leaf on the overall aromatic profile of the lavender essential oil.
