KLF2 regulates neutrophil activation and thrombosis in cardiac hypertrophy and heart failure progression

Summary: The role of inflammation and inflammatory cells, particularly neutrophils, in cardiac hypertrophy and heart failure has been investigated in this study. It was found that neutrophils play a crucial role in the pathogenesis and progression of heart failure by triggering thrombosis in small myocardial vessels through the KLF2/NETosis pathway. Targeting neutrophils, NETosis, or thrombosis can alleviate pathological changes and preserve cardiac dysfunction.

Derivatives of 1-DNJ like miglitol, miglustat, and migalastat have clinical applications in treating diseases such as diabetes and lysosomal storage disorders.

Patients may have variable responses to treatment due to heterogeneity in disease manifestations, and understanding mutation-specific mechanisms may guide therapeutic strategies. Preclinical and clinical studies suggest that a preventative therapeutic approach and consideration of mutation-specific mechanisms may be more efficacious in treating HCM.
