Non-canonical phosphorylation of Bmf by p38 MAPK promotes its apoptotic activity in anoikis

Zhe Zhi et al.

Summary: This study uncovered a new regulatory mechanism of Bmf activity during anoikis and provided a structural basis for Bmf cytoskeleton tethering and dissociation. The activation of p38 MAPK directly phosphorylated Bmf at multiple sites, including T72, which blocked the interaction between Bmf and DLC2. Additionally, a phosphomimetic mutation of T72 was found to enhance Bmf apoptotic activity in vitro and in a knock-in mouse model.

Adapted to Survive: Targeting Cancer Cells with BH3 Mimetics

Joan Montero et al.

Summary: A hallmark of cancer is the ability of cancer cells to evade cell death. The approval of the anti-apoptotic BCL2 antagonist venetoclax has highlighted the potential of targeting apoptotic pathways in cancer treatment. However, responses to pharmacologic modulators of cell death have been inconsistent. This review discusses emerging concepts in the use of these therapies and proposes that changes in apoptotic dependency induced by treatment need to be targeted for precise deployment of these new agents.

The manipulation of apoptosis for cancer therapy using BH3-mimetic drugs

Sarah T. Diepstraten et al.

Summary: Apoptosis is a programmed cell death regulated by BCL-2 family proteins. BH3-mimetic drugs have been developed to target prosurvival proteins in cancer cells. These drugs are designed to induce apoptosis and have shown efficacy in certain cancers but faces challenges like resistance and toxic effects.

BH3 profiling identifies ruxolitinib as a promising partner for venetoclax to treat T-cell prolymphocytic leukemia

Charles Herbaux et al.

Summary: Novel agent classes like JAK/STAT and T-cell receptor pathway inhibitors, as well as HDAC inhibitors, have shown preclinical activity in T-PLL. BH3 profiling revealed that primary T-PLL cells mainly depend on BCL-2 and MCL-1 proteins for survival. The combination therapy of venetoclax and ruxolitinib demonstrated significant efficacy in refractory T-PLL patients, especially in those with JAK3 mutations.

Targeting MCL-1 in cancer: current status and perspectives

Haolan Wang et al.

Summary: MCL-1, an antiapoptotic protein, is frequently overexpressed in many tumor types and closely associated with tumorigenesis, poor prognosis, and drug resistance. Its central role in regulating the mitochondrial apoptotic pathway makes it an attractive target for cancer therapy. Progress has been made towards MCL-1 inhibitors and some have entered clinical trials.
