Multistate Redox-Switchable Ion Transport Using Chalcogen-Bonding Anionophores

Andrew Docker et al.

Summary: Synthetic supramolecular transmembrane aniono-phores are a promising class of anticancer chemotherapeutics, and achieving spatiotemporally controlled activity is crucial for their targeted application. In this study, a series of chalcogen-bonding diaryl tellurium-based transporters were developed, with their anion binding potency and anionophoric activity controlled through reversible redox cycling. This unprecedented in situ reversible multistate switching allows for switching between ON and OFF anion transport, and is crucially achieved with biomimetic chemical redox couples.

Reversible Stimuli-Responsive Transmembrane Ion Transport Using Phenylhydrazone-Based Photoswitches

Manzoor Ahmad et al.

Summary: Phenylhydrazone-based synthetic receptors are reported as stimuli-responsive anion transporters. X-ray crystallographic studies confirmed the preorganized anion binding E-state and closed non-binding Z-state. Detailed ion transport mechanistic studies confirmed CI/anion exchange as the primary ion transport process. Light-stimulated E-to-Z isomerization leads to a substantial decrease in ion transport activity across the lipid bilayer membrane. Reversible OFF and ON activity switching was studied using light and acid stimuli, respectively.

Controlling transmembrane ion transport via photo-regulated carrier mobility

Laura E. Bickerton et al.

Summary: This research introduces a novel mechanism for photo-gating ion carriers, which controls the mobility of carriers within the lipid bilayer membrane to achieve switchable ion transport activity. This approach does not involve direct functionalization of the ion binding site, providing a new avenue for precise control over ion carrier activity.

Stimuli-Responsive Anion Transport through Acylhydrazone-Based Synthetic Anionophores

Manzoor Ahmad et al.

Summary: Photoswitchable acylhydrazone-based synthetic anionophores were reported, showing the attenuation of ion transport activity through a phototriggered E to Z photoisomerization process, with resulting high kinetic stability in the photoisomerized deactivated state due to an intramolecular hydrogen bond. Switchable OFF-ON transport activity was achieved through the application of light and an acid-catalyzed reactivation process.

A Pyridyl-Linked Benzimidazolyl Tautomer Facilitates Prodigious H+/Cl- Symport through a Cooperative Protonation and Chloride Ion Recognition

Abhishek Mondal et al.

Summary: The two pyridyl-linked benzimidazolyl hydrazones are reported as HCl cotransporters with superior efficiency compared to the natural product prodigiosin. They provide an appropriate HCl binding site through cooperative protonation and chloride ion recognition, leading to lysosome deacidification. These compounds induce cytotoxicity towards human breast cancer cells while being relatively nontoxic to noncancerous cells.
