GdIII-19F Distance Measurements for Proteins in Cells by Electron-Nuclear Double Resonance

Manas Seal et al.

Summary: Studies on protein structure and dynamics are usually conducted in dilute buffer solutions, which differ significantly from the crowded environment inside cells. The DEER technique can track protein conformations in cells by providing distance distributions between spin labels. However, distances below 1.8 nm cannot be accessed by this technique. This study demonstrates that Gd-III-F-19 Mims ENDOR measurements can cover part of this short range.

Summary: ENDOR spectroscopy is used to detect nuclear spins and hyperfine interactions. The main challenge is spectral analysis due to broad resonances and a large parameter space. A statistical approach followed by gradient-based fitting procedures is proposed for analyzing experimental 263 GHz 19F ENDOR spectra.

Application of W-band 19F electron nuclear double resonance (ENDOR) spectroscopy to distance measurement using a trityl spin probe and a fluorine label

N. B. Asanbaeva et al.

Summary: In this study, the suitability of high-field ENDOR spectroscopy in the W-band for triarylmethyl and fluorine label pairs was investigated using newly synthesized model compounds. The results showed that strong magnetic fields allowed for distinguishing nuclear frequencies of F-19 and protons with sufficient resolution. The study also applied this approach to investigate the thiol exchange process between triarylmethyl-labeled and F-19-labeled human serum albumin molecules.

19F Electron-Nuclear Double Resonance Reveals Interaction between Redox-Active Tyrosines across the α/β Interface of E. coli Ribonucleotide Reductase

Andreas Meyer et al.

Summary: Ribonucleotide reductases (RNRs) play a key role in DNA replication and repair by catalyzing the reduction of ribonucleotides. In Escherichia coli, RNRs undergo a multistep radical transfer process involving proton-coupled electron transfer. Advanced EPR spectroscopy has been used to study this mechanism and revealed important insights into the behavior of the RNR enzyme complex.

Short-range ENDOR distance measurements between Gd(iii) and trifluoromethyl labels in proteins

Martyna Judd et al.

Summary: This study demonstrates the use of gadolinium ions as paramagnetic tags for measuring longer distances in proteins. Gadolinium ions have greater electronic spin and faster electronic spin-lattice relaxation compared to nitroxide radicals, enhancing the sensitivity of the measurements. The use of trifluoromethyl groups further enhances signal intensity.

Benchmark Test and Guidelines for DEER/PELDOR Experiments on Nitroxide-Labeled Biomolecules

Olav Schiemann et al.

Summary: Distance distribution information obtained by pulsed dipolar EPR spectroscopy plays a crucial role in structural biology research, providing unique constraints for integrative structural modeling. Quality standards for sample preparation, measurements, data conversion, interpretation, and reporting have been defined to ensure the reliability of structural models and biological conclusions, with support from a multi-laboratory benchmark study and analysis of data sets with known distance distribution ground truth. The guidelines mainly focus on proteins labeled with nitroxides and double electron-electron resonance measurements, while also offering suggestions for similar cases.
