In vitro antileishmanial efficacy of antiplasmodial active aminoquinoline-chalcone hybrids

Janine Aucamp et al.

Summary: The overlapping distribution of malaria and leishmaniasis increases the risk for comorbidity and there is a need for new anti-infective drugs to treat these diseases and coinfections.

Indolin-2-one Nitroimidazole Antibiotics Exhibit an Unexpected Dual Mode of Action

Till Reinhardt et al.

Summary: This study reveals a new dual mode of action for antimicrobial drugs. These drugs not only directly inhibit the key enzyme topoisomerase IV responsible for bacterial DNA replication, but also cause damage to bacterial DNA and proteins through the production of harmful reactive species via reductive activation.

Mutations in an Aquaglyceroporin as a Proven Marker of Antimony Clinical Resistance in the Parasite Leishmania donovani

Jade-Eva Potvin et al.

Summary: This study identified gene mutations in AQP1 associated with antimony resistance in Leishmania donovani by sequencing the genomes of clinical isolates and conducting experimental validation through CRISPR-Cas9-mediated genome editing. Restoring the wild-type AQP1 in resistant isolates sensitized them to antimonials, while introducing the mutation in sensitive isolates led to antimony resistance. Through genomic analysis and gene editing, the role of AQP1 mutations in clinical resistance was confirmed.

Synthesis and in vitro antileishmanial efficacy of novel benzothiadiazine-1,1-dioxide derivatives

Daisy K. Mangwegape et al.

Summary: Leishmaniasis is a major parasitic disease affecting tropical and subtropical developing countries, with limited treatment options. Through in vitro experiments with benzothiadiazine derivatives, it was found that these compounds have potential as antileishmanial agents by showing activity against parasites and no toxicity to mammalian cells.

Bio-guided isolation of anti-leishmanial natural products from Diospyros gracilescens L. (Ebenaceae)

Cyrille Armel N. Njanpa et al.

Summary: The study aimed to discover new inhibitors of Leishmania spp. using extracts from Diospyros gracilescens. The hydroethanolic crude extract of D. gracilescens trunk showed the most potent antileishmanial activity, with the hexane fraction showing the most potent activity. Further studies on the isolated compounds could lead to potential drug discovery for visceral leishmaniasis.
