The Kynurenine Pathway and Kynurenine 3-Monooxygenase Inhibitors

Summary: Under inflammatory conditions, the upregulation of kynurenine 3-monooxygenase (KMO) enzyme leads to the production of toxic metabolites associated with various diseases. Developing KMO inhibitors to reduce the production of these toxic metabolites is necessary. The KMO crystal structure provides an opportunity for structure-based inhibitor design. This review discusses the kynurenine pathway, KMO enzyme, and the potential clinical use of KMO inhibitors.

Imbalance in KP metabolism may contribute to the development of psychiatric sequelae after TBI.

New therapeutic strategies based on defined molecular mechanisms and neural circuits may offer promise for improving the treatment of bipolar disorder.
