Improving the Function of Meningeal Lymphatic Vessels to Promote Brain Edema Absorption after Traumatic Brain Injury

Jianwen Liao et al.

Summary: This study investigated the effects of ketoprofen, 9-cis retinoic acid (RA), and vascular endothelial cell growth factor-C (VEGF-C) on brain edema after traumatic brain injury (TBI). The results showed that these substances can improve the function of meningeal lymphatic vessels, promote brain edema absorption, reduce neuroinflammation, and improve neurological function.

Summary: The impact of mTBI on the cerebral vasculature and the potential of CVI as a therapeutic target for improving clinical outcomes are highlighted in this article. Despite limited research in this area, focusing on the cerebrovascular system could lead to substantial improvements in patient care for mTBI.

Treatment with vascular endothelial growth factor-A worsens cognitive recovery in a rat model of mild traumatic brain injury

Mujun Sun et al.

Summary: In a rat model of mTBI, treatment with VEGF-A failed to improve cognitive recovery and even showed negative effects in some cases. Cognitive deficits seemed to be associated with gene expression changes related to neuroinflammation and hypoxia.

The adaptor protein Grb2b is an essential modulator for lympho-venous sprout formation in the zebrafish trunk

Cristina Mauri et al.

Summary: Recent studies have shown that the adaptor protein Grb2b plays a crucial role in lymphatic versus venous sprout cell behavior, with higher requirements for Grb2b and Vegfr3 downstream signaling levels in lymphatic structures compared to vein precursor-generating sprouts.

Functional characterization of the dural sinuses as a neuroimmune interface

Justin Rustenhoven et al.

Summary: The CNS-derived antigens in the cerebrospinal fluid accumulate around the dural sinuses and are captured by local antigen-presenting cells to be presented to T cells, promoting tissue resident phenotypes and effector functions within the dural meninges. The dural sinuses play a critical role as a neuroimmune interface, facilitating the surveillance of brain antigens under steady-state conditions.

Novel Synthetic and Natural Therapies for Traumatic Brain Injury

Denise Battaglini et al.

Summary: Traumatic brain injury (TBI) is a major cause of disability and death worldwide, and the "staircase approach" is considered the only validated treatment method. In addition to conventional therapies, novel neuroprotective therapies have been proposed. Treatment strategies should be based on specific and distinct targets such as brain edema, neuronal activity, anti-inflammatory effects, and more.

New lymphatic cell formation is associated with damaged brain tissue clearance after penetrating traumatic brain injury

Fan-Wei Meng et al.

Summary: The study characterized the tissue repair response after penetrating traumatic brain injury in mice by examining tissue morphology and the expression of lymphatic-specific markers. The findings suggest a clearance mechanism involving lymphangiogenesis, with the appearance of new lymphatic cells in damaged brain tissue. Further research is needed to clarify the origin of these cells and their role in brain tissue repair.

Antimicrobial immunity impedes CNS vascular repair following brain injury

Panagiotis Mastorakos et al.

Summary: Traumatic brain injury and stroke are often complicated by systemic infections, which can impair central nervous system revascularization and repair, potentially leading to chronic disrepair. Viral infections interfere with brain repair by disrupting IFN-I signaling, highlighting a critical impact on recovery.

Skull and vertebral bone marrow are myeloid cell reservoirs for the meninges and CNS parenchyma

Andrea Cugurra et al.

Summary: The meninges contain a pool of monocytes and neutrophils supplied by adjacent skull and vertebral bone marrow, rather than by the blood. Under pathological conditions, CNS-infiltrating myeloid cells can originate from the brain borders and have distinct transcriptional signatures from their blood-derived counterparts. Therefore, myeloid cells from adjacent bone marrow niches are strategically placed to supply innate immune cells under both homeostatic and pathological conditions.
