Palladium-Catalyzed Dearomative [4+2]-Cycloaddition toward Hydrocarbazoles

Summary: An efficient method has been developed for the construction of hydrocarbazoles bearing three continuous sterically hindered stereocenters via a highly diastereoselective palladium-catalyzed [4 + 2]-cycloaddition/ dearomatization of 3-nitroindoles. The multifaceted transformations of the products demonstrate the further synthetic utility of this method. The catalytic asymmetric aspect of this transformation has also been explored.

Oxidative Cyclization of Aryl Ynones with NaNO2 for the Divergent Synthesis of NO2-Containing Spiro[5.5]trienones, Indenones and Thioflavones

Summary: A K2S2O8-promoted nitration/spirocyclization reaction of biaryl ynones using inexpensive NaNO2 as the nitro source has been reported. This cascade reaction involves the nitration of alkynes followed by 6-exo-trig spirocyclization and dearomatization, resulting in the formation of NO2-containing spiro[5.5]trienones in yields of 63-85%. Furthermore, the reaction provides a practical method for the synthesis of nitrated indenones and thioflavones with yields of 58-85%.

Radical spirocyclization of biaryl ynones for the construction of NO2-containing spiro[5.5]trienones

Electrochemical Nitration with Nitrite

The Use of Potassium/Sodium Nitrite as a Nitrosating Agent in the Electrooxidative N-Nitrosation of Secondary Amines

Summary: A new method for the electrochemical N-nitrosation of secondary amines using sodium/potassium nitrite as a nitrosating agent is reported. This approach eliminates the need for strong acid and has good functional group tolerance. The reaction is compatible with late-stage modification of pharmaceutical compounds and shows high reaction efficiency on a gram scale.

Silver-Catalyzed Asymmetric Dearomatization of Electron-Deficient Heteroarenes via Interrupted Barton-Zard Reaction

Summary: In this study, a catalytic asymmetric dearomatization reaction of electron-deficient heteroarenes with alpha-substituted isocyanoacetates was reported, leading to a range of optically active pyrrolo[3,4-b]indole derivatives. The reaction yielded good results with high stereoselectivities, using a catalytic system consisting of a cinchona-derived amino-phosphine and silver oxide. This method features wide substrate scope, mild conditions, and provides a new strategy for developing asymmetric dearomatization reactions.

Synthesis of 3-nitroindoles by sequential paired electrolysis
