Severe malaria in children and adolescents in Southeast Gabon

Summary: This study describes the epidemiological aspects of severe malaria in rural, semi-rural, and urban areas of southeast Gabon. The study found that the epidemiological characteristics of severe malaria were similar in the three settings, but more severe in urban areas. Children under 5 years of age are the most vulnerable age group.

Mapping genetic markers of artemisinin resistance in Plasmodium falciparum malaria in Asia: a systematic review and spatiotemporal analysis

Summary: This study aimed to provide up-to-date spatial distribution visualisations of kelch13 (K13) gene markers of artemisinin resistance in Plasmodium falciparum for policy makers. Data from 72 studies comprising K13 markers from 16,613 blood samples collected from 1991 to 2020 from 18 countries were collated, showing a steady increase in the prevalence of WHO-validated K13 markers over time. Further consistent data collection, over more extended periods, and rapid sharing of data are needed to map the spread and evolution of resistance to inform policy decisions better.

Efficacy of artesunate-amodiaquine and artemether-lumefantrine for uncomplicated Plasmodium falciparum malaria in Madagascar, 2018

Summary: This study evaluated the efficacy of ACT and molecular markers of anti-malarial resistance in treating uncomplicated falciparum malaria in Madagascar. The results showed that both ASAQ and AL had therapeutic efficacies above 90%, with no genetic evidence of artemisinin resistance observed. However, the presence of certain pfmdr1 genotypes was associated with tolerance to lumefantrine.

Association of Plasmodium falciparum kelch13 R561H genotypes with delayed parasite clearance in Rwanda: an open-label, single-arm, multicentre, therapeutic efficacy study

Summary: This study confirms the emergence of artemisinin partial resistance in Rwanda. Although artemether-lumefantrine remains efficacious, vigilance for decreasing efficacy and further characterization of artemisinin partial resistance should be considered. Evaluation of additional antimalarials in Rwanda is also recommended.

Summary: The global spread of drug resistance to antimalarial treatments poses a serious public health risk. Through sequencing samples from therapeutic efficacy studies in Africa, mutations associated with partial artemisinin resistance were detected but no validated or candidate artemisinin-resistance mutations were found.
