Morphological alterations in C57BL/6 mouse intestinal organoids as a tool for predicting chemical-induced toxicity

Summary: Intestinal organoids can be used as an alternative model for toxicity testing, but the relationship between specific morphological changes in organoids and chemical-induced toxicity needs further exploration. In this study, we generated mouse intestinal organoids and analyzed the morphological changes induced by four metals (cadmium, lead, hexavalent chromium, and trivalent arsenic). These morphological alterations were concentration-dependent and correlated with specific intestinal toxic effects. The benchmark dose values of morphological alterations were lower than those of conventional bioassays. We also found that the morphological features of organoids were metal-specific and mediated by different signaling pathways. These findings provide valuable insights into the relevance of morphological alterations in organoids to specific toxic endpoints.

Exposure to copper and/or PVC microparticles resulted in hyperemia, leukocyte infiltration, and an increase in melanomacrophage centers number and area. Generally, the severity of the lesions followed the order: PVC microparticles < copper < copper+ PVC microparticles. In conclusion, PVC microparticles act as a copper vector, facilitating copper accumulation in the fish liver and increasing tissue damage.

Detection of microplastics in human lung tissue using μFTIR spectroscopy

Dietary exposure to polystyrene nanoplastics impairs fasting-induced lipolysis in adipose tissue from high-fat diet fed mice

Summary: The health concerns of microplastics (MPs) and nanoplastics (NPs) have been increasing, but there is still a lack of key indicators to evaluate the adverse risks of MPs/NPs. This study demonstrates that fluorescent polystyrene nanoplastics (nPS) can distribute in white adipose tissue (WAT) of mice and reduce lipolysis under stimulation in vitro and ex vivo. Chronic oral exposure to nPS at relevant concentrations impairs lipid mobilization in obese mice, leading to larger adipocytes in WAT. Additionally, nPS exposure induces macrophage infiltration in the small intestine and increases lipid accumulation in the liver, disrupting systemic metabolism. These findings highlight the potential obesogenic role of nPS via diminishing lipid mobilization in WAT and suggest using lipolysis relevant parameters for evaluating the adverse effect of MPs/NPs in clinics.
