Molecular Plasticity of Crystalline CK2α′ Leads to KN2, a Bivalent Inhibitor of Protein Kinase CK2 with Extraordinary Selectivity

Dirk Lindenblatt et al.

Summary: CK2a and CK2 alpha' are paralogous catalytic subunits of CK2, playing roles in tumorigenesis and viral spread. The alpha D pocket in CK2 alpha' can undergo structural plasticity, leading to the design of the selective bivalent CK2 inhibitor KN2. KN2 binds to CK2 with high affinity, is cell-permeable, and inhibits CK2 activity in cells.

Reduced serum protease activity in Complex Regional Pain Syndrome: The impact of angiotensin-converting enzyme and carboxypeptidases

Simone Koenig et al.

Summary: The study confirmed that the majority of DBK was degraded by ACE and CPN in serum of CRPS patients. Significant differences in serum protein expression were observed only in females younger than 46 years with CRPS as compared to healthy individuals, suggesting potential involvement of defense/immunity-related proteins and the renin-angiotensin system in the pathophysiology of CRPS. Further investigation is needed to understand the impact of CPN in CRPS.
