Development of 18F-Labeled Bispyridyl Tetrazines for In Vivo Pretargeted PET Imaging

This study successfully labeled bispyridyl tetrazines for the first time and demonstrated their application in pretargeted PET imaging. Synergistic Experimental and Computational Investigation of the Bioorthogonal Reactivity of Substituted Aryltetrazines

Summary: Tetrazines have wide applications in biomedical field, including pretargeted imaging. In this study, a kinetic investigation on different tetrazine compounds reacting with trans-cyclooctene (TCO) was conducted. The results reveal that the reactivity of tetrazines in different solvents may not correlate well and tetrazines modified in the 2-position of the phenyl substituent show high intrinsic reactivity towards TCO, which is diminished in aqueous systems. These findings enable the prediction of bioorthogonal reactivity and facilitate the development of substituted aryltetrazines for in vivo applications.

Immuno-PET Imaging of Atherosclerotic Plaques with [89Zr]Zr-Anti-CD40 mAb-Proof of Concept

Traditional strong basic conditions used during F-18 labeling procedures limit labeling of base-sensitive scaffolds. Development of a low-basicity elution procedure allowed for successful S(N)2 F-18 labeling of base-sensitive substrates with improved radiochemical yields.

Summary: The development of highly selective and fast biocompatible reactions for ligation and cleavage in bioorthogonal pretargeting has opened up new avenues for targeted delivery of radionuclides and drugs. This approach allows for increased target-to-background ratios at early time-points, reducing radiation burden to healthy tissue and enabling better imaging contrast or higher therapeutic efficiency. Additionally, triggering cleavage of pretargeted antibody-drug conjugates bioorthogonally represents an emerging strategy for controlled release and locally increased drug concentrations.
