The development of 4-IPP nanospheres allows for targeted delivery and sustained release of 4-IPP, demonstrating potent anti-inflammatory effects and providing a promising therapeutic strategy for rheumatoid arthritis.

Obacunone targets macrophage migration inhibitory factor (MIF) to impede osteoclastogenesis and alleviate ovariectomy-induced bone loss

Summary: This study aimed to explore the therapeutic potential of Obacunone (OB) on osteoporosis and its rudimentary mechanisms. In vitro and in vivo experiments showed that OB suppressed the formation and function of osteoclasts. Mechanistically, OB interacted with macrophage migration inhibitory factor (MIF), attenuating receptor activator of nuclear factor kappa B (NF-KB) ligand (RANKL)-induced signaling pathways. This led to a decrease in the expression level of the master transcriptional factor of osteoclastogenesis, nuclear factor of activated T cells 1 (NFATc1), and its downstream osteoclast-specific proteins. OB also alleviated estrogen deficiency-induced osteoporosis by targeting MIF and inhibiting hyperactive osteoclasts.

MIF is essential to the establishment of house dust mite-induced airway inflammation and tissue remodeling in mice

Summary: Macrophage migration inhibitory factor (MIF) is highly expressed in BALF and serum of asthmatic patients and contributes to asthmatic pathogenesis in mice. This study investigated the role of MIF in a more complex and relevant asthma model using house dust mite (HDM) extract. Results showed that MIF deficiency reduced airway hyperresponsiveness, eosinophil infiltration, mucus hypersecretion, and subepithelial fibrosis in HDM-challenged mice. Additionally, MIF blockade prevented the increase of inflammatory cytokines IL-4, IL-5, and IL-13 in the lungs while preserving CCL11 levels. These findings suggest that MIF may be a promising target for asthma therapy.

Co-immunoprecipitation (CoIP) results indicated direct interaction between MIF and SLC3A2. In vivo experiments confirmed that knockdown of both MIF and SLC3A2 inhibited tumor growth and metastasis via the AKT/GSK-3 beta pathway. MIF and SLC3A2 may serve as potential biomarkers for monitoring colorectal cancer treatment.
