Enhancing Pt(IV) Complexes' Anticancer Activity upon Encapsulation in Stimuli-Responsive Nanocages

Maria Sancho-Albero et al.

Summary: Platinum-based chemotherapy is the standard first-line treatment for various types of cancer, including malignant pleural mesothelioma. This study presents a strategy using a breakable nanocarrier to stabilize, transport, and release a platinum(IV) prodrug, which is activated by the presence of glutathione in neoplastic cells. In vitro and in vivo experiments demonstrate the successful release and activation of the Pt-based drug inside cancer cells, leading to a significant reduction in tumor growth.

Lipidic Formulations Inspired by COVID Vaccines as Smart Coatings to Enhance Nanoparticle-Based Cancer Therapy

Marzia Conte et al.

Summary: Recent advances in nanomedicine have led to the introduction and establishment of nanoparticles in cancer treatment and diagnosis, but their application still faces challenges related to biocompatibility and biodistribution. This paper takes inspiration from COVID vaccines to develop a lipidic formulation as a smart coating for solid-state nanoparticles, aiming to ensure efficient and stable shielding of the cargo. Coated zinc oxide nanoparticles doped with iron show a significant increase in cell viability compared to uncoated nanoparticles, demonstrating the potential of highly biostable and well-tolerated nanoparticles in various therapeutic approaches.

Artificial and Naturally Derived Phospholipidic Bilayers as Smart Coatings of Solid-State Nanoparticles: Current Works and Perspectives in Cancer Therapy

Nicolo Maria Percivalle et al.

Summary: Recent advances in nanomedicine for cancer treatment have focused on using liposomes and extracellular vesicles as carriers to deliver therapeutic agents to tumor cells. Solid-state nanoparticles are also of interest due to their various properties and potential applications. However, the use of nanoparticles in nanomedicine may have limitations such as nonspecific uptake, aggregation in biological environments, and possible immunogenicity. A promising strategy is to use phospholipidic bilayers as a biocompatible shield to effectively encase solid-state nanoparticles, expanding the possibilities of cancer nanomedicine and enhancing therapeutic potential through lipid bilayer engineering.
